| In recent years, with the improvement of people's living standard, the demand for pork becomes more and more serious, so the long distance transport of piglets and fattening pigs becomes more and more general. But subsequently, the transportation, as a kind of stress, brings more and more negative effects on the swine, such as disease, death, etc. Many studies had showed that the negative effects were associated with the disorder of neuro-endocrine-immunity network because of the transport stress. To study the transport effects on immune function, the Erhualian piglets of Chinese indigenous, about 20kg weight, were selected in this study. The influence of stressed serum on peripheral blood T lymphocyte proliferation of normal pigs was determined by MTT assay; the results showed that: the serum dilution at 1:4 and 1:8 had significant difference(P<0.05)between control group and stress group, but at 1:16 dilution, there were no obvious difference (P>0.05) between the two groups.Serum interleukin 1βand interleukin 2 levels were assessed by radioimmunoassay; the results demonstrated the concentration of the two cytokines had no significant difference (P>0.05) between control group and stress group, but as an embodiment of cellular immune function, the increasing tendency of IL-2 level was more than that of IL-1β, demonstrating that the transport of 2 hours can enhance the immune functions in piglets. The expressions of IL-1βand IL-2 mRNA in spleens were determined by RT-PCR, the results showed that: the IL-1βmRNA level had no significant difference (P>0.05) between control group and stressed group; while the expression of IL-2 mRNA in the spleen of stressed piglets was significantly higher than control ones. The above results were coincide and demonstrated that the transport of 2 hours was hardly enough to cause the immunosuppression of the piglets, but it stimulated the compensatory emergency response mechanism to cope with the harmful environment. |
