Cucumber is one of the ten most widely grown vegetable crops worldwide and plays important role in meeting human needs. Along with the increasing life level and export amount, the total grown area and yield of cucumber in our country have been increased year by year. The problem of lack of various cucumber varieties with high quality, especially those fit to grow in dependencies, would be more and more serious as the time going.By now there are many genetic maps in cucumber, it would be the time for integration of molecular linkage maps in cucumber, but there are little of the anchor markers for using map integration. In this study, North China type inbred line S94 and Europe Greenhouse type inbred line S06 were used as PCR DNA template denotes. Totally 146 SCAR markers have been obtained from special primers designed by the sequences of RAPD and SRAP polymorphic bands between S94 and S06. These SCAR markers obtained in this research could be at least 10% polymorphic primers validated among the four typical cucumber materials, which would be used to constructing the molecular linkage map in cucumber.Using molecular to construct the genetic map is the base of mapping Traits, also is an important step of molecular assisted selection. We have gotten more than ten map in cucumber, due to the different background of the cucumber's varieties, we can only solve little problems by using a simple one. According to our country's cucumber breeding selection's requirement, especially the varieties'requirement which growing in dependencies, a molecular linkage map was constructed using F2 progenies with one line S52 (South China type) and the other line S06 (European greenhouse type) as parents by the SSR and SCAR molecular makers. This map contains seven linkage maps, total maker number is 24, and total mean interval is 11.6cM. This work walked a key step for the deeply integration between maps.D (Dull fruit skin) and ss (small spines) traits are identified as polymorphic makers, linked in the 6th linkage map. The results show the two traits are controlled by different single dominant gene, respectively. D was linked with the CMC TN71, the distance is 25.8cM, ss is linked with D, while the distance is 11.7cM.
