| In the research of Pennisetum Rich materials from FAFU and JUNCAO Technology,the morphological markers were studied and the reaction and procedure of RAPD-PCR and 1SSR-PCR are optimized.Through the comparative study between Morphology and RAPD markers,ISSR markers of pennisetum genetic diversity,genetic variation about their relationship between the level of molecules and morphology,different molecular markers were explored.The specificity strip of RAPD and ISSR markers were cut,recycled,priced into SCAR marker successfully.The main results are as follows:The 10 penniseetum cuttings growed up after eight mouths,there are 10 morphological characteristics of 10 penniseetum were analyzed:Natural high,stem height,stem coarse,leaves length,leaves width,Internodes length,Tiller number,Internodes number,leaves number and single root weight.The result indicated that there was high genetic diversity in 10 penniseetums,the coefficient of variation between 9.76%-48.48%.Clustering a.4nalysis of 10 penniseetums showed that witch can be divided into three categories:the first category is generally tall,the second category of blade is generally long and wide,P.purpureum Schum cv.Xijing with different appeamace falls into third category.From 120 random primer,there are 22 primers were chosen and used to analyze 14 Penniseetum by RAPD.A total of 199 out of 258 amplification products were polymorphic,percentage of polymorphic loci was 77.1%.Clustering dendrogram with software NTSYSpc 2.0 divided 14 Penniseetum into six categories:NO.1 category:Pennisetum americanum X P.purpureum;NO.2 category:P.purpureum Schum cv.Purple;NO 3category:P.purpureum cv.Jujun cao、P.purpureum × P.tyhoideum;NO.4 category:Pennisetum purpureum Schum.cv.N51、Pennisetum purpureum Schum.cv.N85;NO.5 category:Pennisetum purpureum Schum.’ Guiminyin ’、P.purpureum Schum cv.Taiwan;the rest formed last category.From 95 ISSR primer,there are 17 primers were chosen and used to analyze 14 Penniseetum by ISSR maeker.A total of 232 out of 256 amplification products were polymorphic,percentage of polymorphic loci was 90.6%.Clustering dendrogram with software NTSYSpc 2.0 divided 14 Penniseetum into five categories:NO.1 category:P.purpureum Schum cv.Purple;N0.2category:Pennisetum americanum X P.purpureum;NO.3 category:Pennisetum purpureum Schum.’Guiminyin’、P.purpureum Schum cv.Taiwan;N0.4 category:Pennisetum purpureum Schum.cv.N51、Pennisetum purpureum Schum.cv.N85;the rest formed last category.The specific fragment of 428bp、405bp、519bp、700bp、242bp and 358bp was amplified by random primer S27、S42、S52、S71、S114 in RAPD and u55 in ISSR,According to their sequences,five pairs primers was designed,named SS27-7、SS42-11、SS52-6-2、SS71-8、SS114-12、U55-2.Four RAPD markers and a ISSR marker transformed into SCAR marker.they can accurately distinguish P.purpureum X P.tyhoideum cv.Reyan NO.4、P.purpureum Schum cv.Purple、P.purpureum Schum.cv.Mott、Pennisetum purpureum Schum.cv.N51、Pennisetum purpureum Schum.cv.N85 from 13 Pennisetum. |
