The research was hot topic in litter size genetics at all times. Placental was temporarily apparatus in mammalian, it weightily to growth of fetus in gestation of sow ,The process which provide nutrition maternal to fetus have two ways. First ,The matter transit with vascular endothelial growth factor of placenta and endometrium.Second,The piglets obtain nutrition by the way of increasing placental surface area,which is more important the fomer way.Placeta were collection and measured on difference generations in Large White in actual locale. In order to disting wished differnence litter size, different placental efficiency was compared. The BF gene and FSHβgene were analyzed in Large White pig using PCR-RFLP and PCR-SSCP. More,Genetic Polymorphism of BF and FSH gene and correlation with Reproductive Trait and Placental efficiency were analyzed too. The sequence of Vascular endothelial growth factor was cloned, and relative expression quantity of VEGF was analyzed. The result indicated that:(1)High placental efficiency supply more 2.55 and 3.15 piglets than low placental efficiency on TNB and NBA, Placental efficiency might have positive effective on litter size.(2)Polymorphisms were detected in the loci of BF intron1-1, FSHβintron1-1.A mutation site locate at 142bp of PCR product were detected,it was C→T transition.And, AA, AB and BB genotypes were found in this population. A 273bp sequence which was a retrotransponsons was inserted among the 6473th and 6474th nucleotide of the gene sequence in FSHβ.(3) Least square means of containing different Placental weight,Placental surface area,Placental length,Birth weight and genotype were estimated with fixed model. Genotype of BB had significant effects on the Placental efficiency and Placental surface area(P <0.01) of BF gene ,AA Genotypes had significant effects on the Placental efficiency, Placental surface area and Placental length(P <0.01) of FSHβgene. And Genotype had no significant influence on Placental weight and Brith weigth.(4)The analysis of association between the polymorphisms of BF intron1-1, FSHβintron1-1 and Placental traits showed that Litter size and Placental efficiency would be increased when the frequency of allele in the locus of BF intron1-1 grew up and the frequency of allele in the locus of FSHβintron1-1 would be increased with the same effect.All analyses can be concluded that:The locus of BF intron1-1, FSHβintron1-1 can be considered as molecular marker for selecting Reproduce traits and Placental traits because associations between their polymorphism and reproduce traits had been observed.
