240 one-day-old Sichuan white geese were divided randomly into three groups,and fed on diet with Cu of 10.24mg/kg in control group,Cu of 400mg/kg in copper toxic groupâ… and Cu of 600mg/kg in copper toxic groupâ…¡to study effect of copper toxicity on organs and some blood parameters in geese for 40 days,respectively.The expression of Ctr-1 gene mRNA in geese liver of different groups were studied by the method of RT-PCR at the end of test,to study the pathogenesis of copper intoxication in geese.Two groups of copper intoxication showed distinctive clinical signs,and the death were up to the highest pea after about 30 days.The results showed that,copper toxic was sensitive to geese about 30 days.The incidence of copper toxic groupâ… was 50%,and copper toxic groupâ…¡was 100%,the mortality in the two groups could respectively reach 30%and 80%at the end of test.The research of pathomorphology showed similar lesions in the two copper toxic groups when compared with control group.The cuticle of gizzard appeared thickening and crack;The digestive tract was full of the black-brown or copper-green substance,The intestinal mucosa was swelled and flush,and the submucosa showed modified,necrosis and exfoliation; Histopathologically,the hepatocellular,renal tubules and cardiac muscle mainly showed granular and vacuolar degeneration;Results compared with control group,the content of copper in liver and blood was increased remarkably,respectively;Changes of Haematological Indexes showed that red blood cells in copper toxic groupâ… and copper toxic groupâ…¡became deformed,degenerated and were enlarged or shrinked,or/and necrotic.The populations of red blood cells,hemoglobin contents were much lower than the control group.In addition,the serum Lactate Dehydrogenase(LDH), glutamic-oxaloacetic transaminase(GOT),glutamic pyruvic transaminase(GPT)and malondialdehyde(MDA)activities markedly increased,and the serum Copper-zinc superoxide dismutase(CuZn-SOD),Ceruloplasmin(CP),Glutathione synthetase(GSH-Px)activity markedly decreased.Study of the expression of Ctr-1 gene mRNA in geese liver showed that the two copper toxic groups higher than the control group remarkably.The results showed that,different dosages(400mg/kg and 600mg/kg)of copper can be made successfully chronic poisoning and acute poisoning models.Clinical pathological damage of poisoning geese were markedly on 30 days of experiment.The target organs of copper toxic injury were digestive Tract liver,kidney and heart.The content of store copper in liver and in blood of geese was tended to increase with the ingestion of feed in high concentration copper,It could be a specific index for the diagnosis of copper poisoning.Fed poisoning diet,the serum GOT and GPT activity increased,It showed the serum GOT and GPT were good index to the degree of liver damage.The activity of serum CuZn-SOD,CP and GSH-Px decreased influence antioxidant activity. It could lead to free radicals accumulated in vivo,the membrane system and structure damaged,the brittleness oferythrocyte increased lead to structure damaged and function decreased,and resulted in cells degeneration and necrosis to dysfunction.The Ctr-1 gene mRNA expressive quantity in liver both of poisoning group were tended to increase with the ingestion of feed in high concentration copper,the concentration of copper in cells increased resulted in copper toxic deepen,Eventually, organ dysfunction and failure to death.
