Gladiolus is one of the five most famous cutting flowers in the world, which was with its colorful, flower-shaped unique and long-lasting in water, it likes by people very much. With the development of ornamental plant's tissue culture technology day by day, The study of gladiolus about rapid propagation of tissue culture ,detoxification and transgenic technique become more and more depth. The establishment of quick and simple system for regeneration in gladiolus laid a good foundation for further research into the transgenic. This study was carried out as a focus of a series of tests, to establish a way of regeneration in gladiolus quickly and easily. In the study, we explored and analyzed the factors of different varieties of Gladiolus in the induction, differentiation and subculture in other various stages of growth at first, and then analyzed the factors of the same species in different explants about induction, proliferation and rooting.1. The regeneration in gladiolus established with the cormel in vitro culture: Comparison of the results of lateral bud about four different varieties of gladiolus cormels in the same induction medium, we found that the incidence of induction about lateral bud of P and B are bigger than J and X.. Four species in the bud proliferation medium MS +0.5 mg / L 6-BA +0.1 mg / L NAA could proliferate and subculture normally, the multiplication coefficient of B and P are higher than J and X.2. The regeneration in gladiolus established with inflorescence: With the same species (P) in different parts of the inflorescence for explants, different medium for tissue culture, the result indicated that the differentiation of bisexual pistil was not obvious and the petal was not differentiation completely, the pedicel and receptacle could be induced to callus, and the pieces of the floral axis in the medium of MS+1mg/L 6-BA+1mg/L NAA could induce multiple shoots directly.3. After the bud inducted by stem, to receive the bud on the medium which added different hormone concentration. The statistics is that, The proliferation effect of multiple shoots'subculture which cultivated in the medium of MS +0. 5 mg/L 6 - BA +0. 1mg/L NAA was the best. The optimum medium for rooting was composed of MS+0. 1mg/L NAA.4. The plantlet growth and rooting in the mixed media was better than the only substrate. The feature of the study is that two different ways of the establishment of regeneration system can skip the step of callus induction. The experiment which was used a simple and convenient method, with explants directly inducted to buds, enhanced the speed of establishment of regeneration system greatly. The two different ways provided more choices for the rapid propagation of tissue culture and transgenic of gladiolus.
