Through the basic research of thenonionics nanoemulsion nanotechnology, we handled the rule of nanoemulsion form, and screening the best material and formula to prepare the medicinalem nanoemulsion. We prepared the Furacilin Combination Nanoemulsion by this foundation, and its quality stability, security, release properties and pharmacodynamics were systemically studied.According these,we provid the theory proof for the veterinary clinic .1: Study on the blank nanoemulsion carriers.The Tween80 and RH40 et al. were used as surfactant, the propylene glycol, Span80 were used as cosurfactant, the oleum olivae, isopropyl myristate and Mineral oil were used as oil. The formulations of the nanoemulsion was optimized by studying the pseudotertiary phase diagrams, the factors of influencing nanoemulsion formation were investingated such surfactant, cossurfactant, oil, etc. The experimental results indicated the formation of nanoemulsion was dependent on the surfactants, the nature of oils and cosurfactants ,the weight ratio of surfactant to cosurfactant and the type of additives in the water phase. The four nanoemulsions systems of Tween80/Span80/IPM/water,RH-40/IPM/water,Tween80/Span80/ Mineral oil /water and Tween80/span80/ oleum olivae /water(Km=9:1,8:2) were limpidity and stability.The average diameter of the sample was below 100nm, The Drop shape and particle size were investigated by transmission electron microscope. which accorded with that of nanoemulsion.2: The advanced development of Furacilin Combination Nanoemulsion.The formulations of the proanthocyanidins nanoemulsion was optimized by studying the pseudotertiary phase diagrams, the effect of various elements on the formation of the nanoemulsiom was investigated. Stability of proanthocyanidins nanoemulsiom was studied. The Ultraviolet absorption wavelength, standard curve, recovery and accuracy were studied by Ultraviolet spectrophotometer, The Furacilin has the maximal absorption in 375nm. The standard curve was A= 0.084C+0.0491(n=6),r=0. 9992.The average recovery is 98.82%. The good linear range was 10~30μg/mL and the RSD is 1.5%. it looked stramineous﹑limpidity and very stable. Electron microscopy presented of the proanthocyanidins nanoemulsiom as small spherical drops with a diameter between 1~100nm. 3: The safety evaluation on Furacilin Combination NanoemulsionThrough the Uterine irritability test﹑hystera acute toxicity test﹑hystera long toxicity test and cell toxicity test to evaluate the safty of Furacilin Combination Nanoemulsion. The result showed that the Furacilin Combination Nanoemulsion has no toxicity and stimulation in the hystera of Mus musculus albus.The cytotoxicity in L929 collagenoblast﹑stroma cell and endometrium cellula epithelialis of Mus musculus albus was all less than 2 grade, the whole have no overt toxicity. So the Furacilin Combination Nanoemulsion has no overt toxicity and fit for the requirements of nanoemulsion administer system.4: The efficacy evaluation on Furacilin Combination NanoemulsionThe efficacy evaluation on Furacilin Combination Nanoemulsion by the bacteriostasis test in vitro of the main endometritis pathogenic bacteria and by the therapeutic test of Mus musculus albus endometritis modle . The result showed that the bacteriostatic action in vitro on E.coli, S.aureas, S.agalactae,Sam onellal, Furanillin combination Nanometer Microemulsion was 3﹑4﹑32﹑3 times than Furanillin dusts.The effective power and cure rate on Mus musculus albus endometritis modle was 100%and 91.7%,much stronger than convention praeparatum,the distinction was most significant(p﹤0.01). Therefore, through in vitro and in vivo model Pharmacodynamic study, the Furacilin Combination Nanoemulsion is a highly effective antimicrobial agent.5: The curative effect of Furacilin Combination Nanoemulsion on the endometritis of cowTreated the sick cows ,the Furacilin Combination Nanoemulsion group as the test group ,the Gong yanqing as positive control group, the cure rate was 88.9%,the effective power was 100%.Compared with the control group, the distinction was significant(p<0.05).The results shows that Furacilin Combination Nanoemulsion has much batter on curative effect of endometritis of cow. In short, This study developed the Nanoemulsion, which is more effective than conventional agents in the treatment of cows endometritis , It is an efficient, safe and stable Nanoemulsion preparations and provide a basis for the clinical treatment of cows endometritis.
