Due to more and more agricultural trades between Cross-Strait, the transport of agricultural product from Taiwan province to Mainland. China may cause a high risk of pest outbreak in the mainland area. It is a challenge for quarantine works and thus the lack of pest risk early-warning, pest risk analysis, rapid detection technique and pests treatment technique couldn't meet the requirements of the agricultural trade. Therefore, the improvement of quarantine ability to prevent the risk form Taiwan province is necessary. The overall goal of this dissertation research is developing the pest risk analysis for Cruciferae vegetable from Taiwan province and establishing a rapid detection method for Tomato Spotted Wilt Virus(TSWV) which is distributed in Taiwan.Initially, the information of Taiwan Cruciferae vegetable pests was collected and thus a main pests list was determined. Based on the condition of distribution, government control, carried possibility and potential economy impact, a Potential Quaratine Pests (PQP) list was screened from main pests list. Then, the risk level was evaluated and there were 6 Quarantine Pests (QP) and 2 Regulated non-Quarantine Pests (RNQP) were confirmed. The uncertainty of pest risk analysis was mentioned and some pest risk management measure was proposed.A rapid detection method of Tomato Spotted Wilt Virus(TSWV) was established by using Immunocapture Reverse Transcription Polymerase Chain Reaction (IC/RT-PCR) technique . Ten pairs of primers, based on the conserved nucleotide sequence of nucleocapsid protein gene of different TSWV strains were designed and synthesized and a pair of specific primers was screened. IC/RT-PCR was conducted by using this specific primer. IC/RT-PCR product was according with the anticipation. Sequence analysis indicated that there is 99 % homology between the IC/RT-PCR product and the published virus sequence (No. X61799.1 in Ncbi Datebase). And there are some other problems still need to be studied.
