With the electrophoresis Technology development, the application of capillary electrophoresis, on the one hand, the detection of molecular markers based on PCR became rapid, sensitive and efficient; on the other hand, it need a higher quality genome DNA. Maize (Zea mays L.) as a one-year-old grass, have high contents of polysaccharides, polyphenols, or other secondary metabolites that can interfere with DNA extraction and purification. In this research, we investigate to extract DNA from seedlings, young leaf, matured leaf, corn seed etc, in attempt to establish an optimal system of high-quality genomic DNA isolation, purification, quantification that is free of sample-date and materials. This will provide a technical basis for maize cultivars' identification, establishing DNA fingerprinting pool, also giving a widespread application of maize cultivar protection, marker-assisted selection in breeding and many molecular downstream researches. The results were as follows:1 Using alkalinelysis method, improved SDS, improved CTAB, high-salt low-pH method, DNA extraction kit extract DNA from maize young leaf, improved SDS, improved CTAB, high-salt low-pH method had made some certain simplification compared with the original method, OD260/OD280 is 1.7-1.9, the DNA quality was still good, can be applied fluorescent primers SSR-PCR and capillary electrophoresis Detection.2 Based on high-salt low-pH method, some treatment with different sampling, purifying, precipitating were studied to preserve the effect on genomic DNA extraction in corn. The results indicated that as the tissue developed, the yield of DNA decreased. (2) DNA extracted from seeds is of poor quality , because the seeds contain more polysaccharides, proteins, fats. In addition DNA yield is less.
