PCR-SSCP technique was applied in this study that was designed to investigate the effect of PRKAB2 gene on broiler meat and carcass traits.The broilers from Sichuan Daheng Poultry Breeding Company and Animal Science Academy were used in this research.The research from three-site to a PCR-SSCP genetic polymorphism of PRKAB2 gene,combining to determination of the production means,analyzing the relativity of different genotype and slaughtering and meat quality character,attemptting to provide chicken breeding molecular markers.Designs three pairs of primers in three different exons,and identifies three locuses.PRKAB2 gene to a PCR-SSCP detection and integration of the production means, and analysis the hehedity polymorphism the results of the trial show that:in the eight strans,the five homozygosis strains belong to midrange polymorphism,so which have potentiality to select.In the three hybrid strans,the gene distribution is in line with the distribution of Hardy-Weinberg equilibrium,we can see that in the breeding process,the direction of the crossing stains is highly,the effection is obvious.Combining to determination of the production means,analysis the differences of three locuses between the genotype in the slaughtering and meat quality character.Through theleast-squares analysis andsignificant test,discovers that the PRKAB2 gene in the three sites each genotype on the slaughter of live weight,carcass weight and abdominal fat weight and percentage of abdominal fat the impaction reached a significant level(P<0.05).In addition, it also shows that,PRKAB2 gene locus 1 on the thigh muscle weight and leg muscle rate impaction are also significant(P<0.05),the additive effection is13.32 g and 1.25 percent. However,meat quality character,the AA homozygous genotype of all three-sitemuscle fiber density are more significantly higher than BB(P<0.05).The gene can significantly improve the content of intramuscular fat(IMF).The impaction of PRKAB2 gene locus 1 and 3 on the fibre diameter is significant(P<0.05).AA-genetype of muscle fiber diameter are significantly higher than the BB and AB-genotype(P<0.05),the additive effection of their muscle fiber diameter were 1.300 and 1.260.These results show that,PRKAB2 gene and the chicken slaughter traite and meat quality traite are correlative,which can raise the quantity of meat and improve the qulity of meat.It is probably a selected gene affecting chicken slaughter traite and meat quality trait.
