Wheat is one of the most important crop in world, which has the highest yield and crop area. The goal of high production is the breeder pursues inceasingly, which is also demanded by the entire society of food security significantly. wheat heterosis which was considered one of efficient resistance paths and raising the output of wheat .Two-lines system is the hot spots which wheat heterosis study at present. The photoperiod-thermo sensitive male sterility line is core material of two line system in hybrid wheat breeding .To research the genetic basis of quantitative traits and identify the male-sterility and red-auricle genes in wheat, An DH population with 120 lines derived from cross between BS210 and O201 was sown with two replicates in Beijing and Fuyang, Anhui Province . SSR markers had been screened the polymorphic between BS210 and O201.The rate of fructification investigation was carried out before harvest during 2005~2006 and 2006~2007 cropping seasons. Photoperiod-thermo sensitive male sterility and red-auricle genes had been mapped using SSR markers, BSA and QTLs . The results are as follows:(1) 1188 SSR primers had been screened the polymorphic between BS210 and O201,amang which 202 SSR primers have polymorphism, and detected the segregation in DH lines using polymorphic SSR markers.Genetic linkage map had been constructed by QGA_CN software.the genetic map contains 21 chromosomes, comprised 147 loci and 2600.1cM with a mean marker interval of 17.7cM.(2) DNA pools had been bulked.202 primers were used for the detection of polymorphism.Five markers had polymorphic amplification products between fertile and sterile pools. Two markers had polymorphic amplification products between red and green pools by PCR amplification.The result showed that,the male sterility genes were located on 2A,2B,6B,4A.the red-auricle genes were located on 3B.(3) QTLs had been detected by WinQTLCart software through composite interval mapping analysis.The result showed that: when LOD>2.5, the relative male-sterility gene of wheat which was detected were located on the 2A,2B,3A,7D. LOD scores 2.5 to 5.0,and accounting for 14.0-30.3% of the phenotypic variance. The relative red-auricle gene of wheat which was detected beween the marker WMC182 and WMC54 was located on the 3BL.Coupled with the result of BSA ,the male-sterility genes were located on chromosome 2AS,2BL and 3AL,the interval markers were between Xgwm1053 and Xwmc177, Xwmc474 and Xwmc592 , Xwmc265 and Cfa2262, accounting for 23.9%, 28.3%,and 30.3% of the phenotypic variance. The distance to the nearest marker is 19.1 cM,2.8 cM and7.5cM. The red-auricle gene was located on the 3BL, accounting for 24.1% of phenotypic variance,the distance to the nearest marker WMC182 is 7.1cM,the red-auricle gene derive from BS210.(4) The interactional map of QTLs had been constructed by QTLNetwork software. The result indicated that ,the interaction widely exists between the QTLs. the main QTLs are influenced by other QTLs and existing the interaction btween the QTLs and environment,which controll the wheat fertility character.
