Soybean mosaic virus(Soybean mosaic virus,SMV)is an important soybean infecting agents that leads to leaf discoloration and malformation,plant dwarfism and seedcoat mottling.This results in severe yield loss and affects soybean appearance quality.In this study,soybean seeds were collected from main soybean producing regions of the United States and Heilongjiang,china.Seeds from Songxi,Fuzhou and Zhangzhou of Fujian province and Wendng of Shandong province, where outbreak of Soybean mosaic disease have been reported,were also collected. The viruliferous percentage and rate of emergence of the seeds were detected.The symptoms of the seedlings were compaired.ELISA indicated that viruliferous percentage of the seeds were 12.5%(SMV-MG),15.63%(SMV-ZZH),9.38% (SMV-FZH),11.46%(SMV-SX),10.42%(SMV-WD)and 11.46%(SMV-DB) respectively.The emergence rate of the seeds were 70%(SMV-MG),50% (SMV-ZHZH),82%(SMV-FZH),60%(SMV-SX),68%(SMV-WD)and 62% (SMV-DB).The priliminary results demonstrated that there was an inverse corelation between the viruliferous percentage and emergence rate of the seeds.No significant differences in the disease onset time of the six varieties were observed, wheareas they differred greatly in symptom appearence.The capsid gene of SMV from the six varieties were amplified using RT-PCR and ligated to the pMD-18T vector.The genes was sequenced after recombant plasmids double digestion experiments,which varified the accuracy of the ligation.Sequnce analysises showd that the genes cloned were highly homologous to CH J.With a referrnce to the reported sequencs of SMV CP gene,phylogenetic trees were constructed using Neibor-Joining and Maximum Likehood methods.Disease investigation showed that occourence of SMV in Zhangzhou of Fujian province,where sorbean has been planted at the large scale,was morderate.ELISA experiments showed that the rates of infection were 12%in Dongyuan town of Longhai,10.5%IN Shiliu of Zhangfu,and 15%in Wuzhai of Heping.
