| There are many kinds of virus which can infect soybean. Researches on the composition and distribution of them in soybean production areas have been reported. Soybean mosaic virus(SMV) is a global soybean disease. It affects yield and seed quality in soybean seriously. Studies in recent years showed that variation of SMV strains in Hangzhou region may have occurred. Using the same SMV strain identification system with YangQinghua(2009) and LiKai(2010),41SMV specimens from HangZhou was identified once more and dynamic monitoring to know the popular strains and their changes trend in this region; At the same time, specimens collected from Anhui province and HeiLongjiang province were tested by DAS-ELISA of12virus to know the virus’kinds and distribution; Alignment of nucleotide sequence of SMV CP gene was done, trying to explore the relationship between homology of CP genes sequences and the resources of SMV isolates or pathogenic reaction. The main results are given as follows:1. Specimens collected from HangZhou in2009were purified with half leaf of "Top-Crop"and obtained15isolates, these isolates were tested by DAS-ELISA and12of them were positive, they were inoculated to10different hosts to identify the SMV strains classification. Among them, isolates20-1ã€2-4ã€17-4belong to SC15strain, and the others belong to SC9strain, therefore, we presumed that SC9and SC15were the predominant strains in HangZhou;2. Specimens collected from Anhui province and HeiLongjiang province were tested by DAS-ELISA, we found that the positive percents of SMV is the highest, they are19.71%and11.68%respectively, so SMV is the main soybean disease in the field. Beside SMV, PSV, SBMV have a higher ratio in Anhui province,14.04%and13.91%respectively. Kinds of virus is the most in XinMaqiao town of Anhui province, there are5PSV positive specimens in TaiHe, there are3SBMV positive specimens in ChangFeng%GuZhen and TaiHe; but in HeiLongjiang province, AMVã€and TNV have a relative higher ratio;10.53%and10.16%respectively. Kinds of virus is the most in BeiAn of HeiLongjiang province, there are2SMV positive specimens in BeiAnã€SuiHuaã€HuaChuan and JiXi, there are3AMV positive specimens in ACheng and HeiLongjiang nongkeyuan, there are2TNV positive specimens in HaiLinã€HeiLongjiang nongkeyuan and A Cheng;3. CP gene sequences of16SMV isolates from china were determined and phylogenetic analysis to exploring the relationship between similarity of CP genes sequences and the resources of SMV isolates or pathogenic reaction. Results showed that:SMV isolates from china were mostly clustered in more closely phylogenetic branches. Their CP gene were highly similar and there are similar isolates home and abroad. There are not directly relationship between sequence similarity of CP genes and symptomatic reactions in differential hosts, at the same time, having some relationships with virus’ sources but not ambiguous. |
