| The aim of this thesis is to solve the shortage problem of Rhodiola resources, studied systemically the influence of planthormone on callus induction and subculture in Rhodiola quadrifida, the hairy roots reduced of different Rhodiola, the effect of Dark induced on micropropagation of Rhodiola fastigiata and optimized cryopreservation process by selecting different parameters. The following results were gained:1) The seeds were good explants of callus inducing. The optimum medium for callus inducing was MS + 6 - BA 3.0 mg/L + NAA 2.0 mg/L + 2,4 - D 1.0 mg/L, and the highest induction rate reached 87.1%.2) The callus subculture was achieved on MS medium with 6 - BA 3.0 mg/L, NAA 1.0 mg/L, and 2,4 - D 1.0 mg/L. Cell culture cycle was 30 days, and the highest biomass level reached 13.72 gDW / L on day 21.3) During the inducing process of Rhodiola hairy root, except the Rhodiola Rose callus cells in vitro induced hairy (but it has also died in the latter culture process), other materials were death due to Browning and pollution.4) The leaf, petiole and stem which fetched from the aseptic seedling of Rhodiola fastigiata as explant and employing different dark induced time, the experimental result indicates the best material is the blade, the optimal time 15 days, in which the buds total amount is 1520 wich is 2.46 times than not cultivated darkly.5) Having investigated the change of the endogenous hormone of Rhodiola fastigiata in the different dark induced process, the result indicates proper dark cultivation heightens the level of endogenous hormone, compared with uncultivated darkly, dark cultivated for 15 days, its endogenous hormone ZT, GA3, the content of IAA and ABA has been improved by 15.6% respectively, 4.9%, 25.2% and 8.2%.6) Having studied the plain change of chlorophyll and cyanine of Rhodiola fastigiata in the different dark induced process, the result indicates proper dark cultivation helped to reduce the plain accumulation of cyanine, raise the content of the chlorophyll. Compared with uncultivated darkly, the plain content of cyanine with dark induced has been reduced by 3.71 times; The content of chlorophyll has been improved by 4 times.7) Aseptic bud of seedling sharp injuring organization of Rhodiola fastigiata as material for ultra-low temperature, results show best outside plant body was the wounded organization, after the ultra-low temperature preservation, the revival rate and survival rate were 56% and 39%, respectively, while the sharp organization of bud were very low, only 29% and 21%, respectively.8) During the process of optimizing the ultra-low temperature preservation, the optimal explant was callus, the optimal cane sugar concentration was 0.3M, The best loading time was 40 min, best dehydrated time 60 min, it was the 40 degrees Centigrade to unfreeze temperature bestly, the survival rate and revival rate of the material were 62% and 58% respectively after ultra-low temperature was kept. |
