Acupuncture,a technology which can increase pain threshold by neurohumoral regulation,is characterized by safety,convenience and less side effect.However,there are still some shortcomings of accupuncture anesthesia such as incomplete analgesia, muscular tension and reaction to visceral pulling.In orde to overcome these shortcomings,some researchers advocate to use acupuncture balanced anesthesia,this anaesthetic method reserves the advantages of acupuncture and overcomes those shortcomings of accupuncture anesthesia,and can be used in some complicated operations.Xylidinothiazoline is anα2 adrenoceptor excitomotor.It has been shown to produce sedation,analgesia,mitigate and relax muscles.But when it was used on ruminants,the anesthetic level is alaways difficult to control and can cause regurgitation and aspiration of ruminal contents or saliva into the lung.In order to find an optimal anesthesia method which can be better applied in ruminants,anesthetic effectiveness of electro-acupuncture combined with xylidinothiazoline on anesthesia and its effect on immune function in goats were observed.Thirty hybrid goats were randomly divided into five groups,including xylidinothiazoline 0.5mg/kg group,xylidinothiazoline 2mg/kg group, electro-acupuncture group(EA group),electro-acupuncture combined with xylidinothiazoline 0.5mg/kg group(EA with xylidinothiazoline group) and control group.Stimulation of "baihui santai" and "ergen sanyangluo" points with 36Hz and 1.0~1.5V induced anesthesia in EA-treated groups,EA lasted for 120min.Xylidinothiazoline gived intramuscularly in Xylidinothiazoline-treated group.The pain threshold,respiration rate(RR),heart rate(HR),mean arterial pressure(MAP) and body temperature were observed at 0,5 25,45,65 and 95min after xylidinothiazoline injection or at 0,30,50,70,90 and 120min after electric-acupuncture stimulation.Venous blood samples were taken at 0,1.5,24,48, 96 and 168h after the drug injection(or 25 minutes induction of anaesthesia). erythrocyte-yeast rosette rate(RYRR),leukocyte phagocytosis rate the lymphocyte proliferative rate and plasm cortisol concentration were measured,respectively.The pain threshold in EA with xylidinothiazoline group was not significanly different from that in xylidinothiazoline 2mg/kg group,but was significantly greater than that in xylidinothiazoline 0.5mg/kg group or EA group.There was no signficant difference in RR between EA group and EA with xylidinothiazoline group. Xylidinothiazoline 0.5mg/kg and 2mg/kg reduced RR significantly after the drug injection.The HR suppressed significantly in EA with xylidinothiazoline group,xylidinothiazoline 2mg/kg and xylidinothiazoline 0.5mg/kg group after the drug injection.The HR in EA with xylidinothiazoline group was significant greater than that in xylidinothiazoline 2mg/kg or 0.5mg/kg group at 5,25,45 and 65 min after the drug injection,but significantly less than that in EA group at 5,25,45,65 and 95min after the drug injection.The body temperature decreased significantly in EA with xylidinothiazoline group,xylidinothiazoline 0.5mg/kg group and xylidinothiazoline 2mg/kg group after the drug injection,It increased significantly in EA with xylidinothiazoline group compared with xylidinothiazoline 2mg/kg group at 5min after the drug injection,but there was no significant difference in body temperature between xylidinothiazoline 0.5mg/kg group and EA with xylidinothiazoline group.There was no signicicant difference in RYRR,leukocyte phagocytosis rate and lymphocyte proliferative rate between EA with xylidinothiazoline group and control group.The RYRR in xylidinothiazoline 0.5mg/kg group at 24h,48h and in xylidinothiazoline 2mg/kg group at 1.5h,24h,48h after the drug injection was significant less than that in EA with xylidinothiazoline group.Leukocyte phagocytosis rate decreased significantly in xylidinothiazoline 2mg/kg group at 24h and 48h after the drug injection campared with EA with xylidinothiazoline group, but was significantly greater than that in EA group.However,there were no significant differnece in leukocyte phagocytosis rate between EA with xylidinothiazoline group and xylidinothiazoline 0.5mg/kg group.The lymphocyte proliferative rate decreased significantly at 24h and 48h after the drug injection in xylidinothiazoline 2mg/kg group,but significantly increased in EA group compared with EA with xylidinothiazoline group,respectively.The plasm cortisol concentration increased significantly in EA with xylidinothiazoline group at 1.5h after the drug injection,and increased significantly in xylidinothiazoline 2mg/kg and 0.5mg/kg group at 1.5h and 24h after xylidinothiazoline injection.Inversely,it decreased significantly in EA group at 1.5h and 24h after the drug injection.Theses results indicate that anesthestic effectivenes in goats induced by EA with 0.5mg/kg xylidinothiazoline is equal to xylidinothiazoline 2mg/kg,but the dosage of xylidinothiazoline is reduced by 75%.The balanced anesthesia in goats have less effect on physiological function and immune function which were measured. Therefoe,EA with xylidinothiazoline is a safe and effective anaesthetic method.
