Acupuncture analgesia is a method to prevent and treat pain, which began to be used on veterinary clinical and gradually developed in the late 1960s. Compared with general anesthesia, acupuncture analgesia has many advantages such as less interference with the physiological, higher resume speed and simpler electro-acupuncture anesthesia equipment. For different kinds of animal with different points, acupuncture analgesia can be used in different operation, such as caesarean section, laparotomy, abomasum incision and hernia repair. In order to understand the analgesic effect of acupuncture, the doctor did a lot of researches in rats, rabbits and other animals. They had a certain degree of understanding in neural mechanisms, neurochemical mechanisms and pathways of acupuncture analgesia. However,the mechanism of acupuncture analgesia for ruminants is unknown. Why the effects of acupuncture analgesia on ruminants is better than other animals? Is it due to the types of neurotransmitters, or different analgesia nucleus in central nervous system, or different nerve conduction pathway. As a novel and more practical method to study neural pathways proto-oncogene have been extensively applied to study the mechanisms of acupuncture analgesia. The experiment is a theoretical basis to study neural physiology, neural pathways and drug research in ruminants acupuncture analgesia.100Hz electroacupuncture stimulation is used on goat's "Santai" and "Baihui" point for 30min. the results of pain threshold determine the analgesic effect. Reperfuse after electroacupuncture stop for 1.5 hours and use SABC immunohistochemical method stained to the production of paraffin sections. Observe the number and distribution of the fos protein in the main analgesic nucleus(habenular nucleus, hypothalamic arcuate nucleus, Ventromedial hypothalamic, paraventricular nucleus, periaqueductal gray, dorsal raphe nucleus, red nucleus, locus coeruleus, parabrachial nucleus, raphe magnus nucleus, gigantocellular reticular nucleus, rostral ventrolateral reticular nuclear, and spinal cord dorsal horn ) in thalamus, hypothalamus,brain stem and other parts. Based on the results we could analyse which nuclear was activated, the possible pain pathway.and compared with the research in rat nucleus.Pain threshold of electro-acupuncture group at 10 min and 50 min had no significant differences with control group, but significant at 30min. In control group, habenular nucleus, hypothalamus arcuate nucleus, red nucleus, dorsal raphe nucleus, locus coeruleus, parabrachial nucleus, gigantocellular reticular nucleus, spinal cord dorsal horn did not have expression of Fos-positive. In habenular nucleus, paraventricular nucleus, arcuate nucleus, periaqueductal gray, dorsal raphe nucleus, locus coeruleus nucleus, parabrachial nucleus, gigantocellular reticular nucleus, raphe magnus nucleus of electro-acupuncture group the expression of Fos-positive cells compared with the control group had significant differences, but in the spinal dorsal horn, red nucleus, ventromedial hypothalamic and rostroventrolateral reticular nucleus expression had no significant.The experimental results showed that, using 100Hz electro-acupuncture to stimulate the goafs "Santai" and "Baihui" acupuncture points, habenular nucleus, paraventricular nucleus, periaqueductal gray, dorsal raphe nucleus, locus coeruleus nucleus, parabrachial nucleus, gigantocellular reticular nucleus and raphe magnus nucleus are involved in the role of acupuncture analgesia. Electroacupuncture analgesia is different with noxious stimulation, and there is no stress on the impact of goats.
