Studies On Macleaya Cordata Alkaloids Extraction & Separation And Application

Macleaya cordata is Chinese traditional herbal drug with plenty of alkaloids.In this paper,the extreaction,separation and active of Macleaya cordata alkaloids were studied,the results as following:1,The extraction method study of Macleaya cordata total alkaloidsThe Macleaya cordata total alkaloids was extracted by ethanol and hydrochloric acid,then to compare this two methods.the following results were obtained:acquired of alkaloids was equal in this two methods,but some constituents can be extracted by ethanol that hydrochloric acid can not,The humbler of constituents extracted from Macleaya cordata by hydrochloric acid was better than extracted by ethanol,and the acquired of Sanguinarine,chelerythrine by hydrochloric acid were better than by ethanol so.Optimal conditions for extraction the total alkaloids from macleaya cordate was studied by hydrochloric acid.The working parameters including extracting temperature,hydrochloric acid content,the ration of material to solvent and extracting time were studied.The best working parameters for extracting macleaya cordate total alkaloids were:extracting time was 4 hours,extracting temperature was 80℃,the ration of material to solvent was 1 to 50,the hydrochloric acid content was 1.5,the macleaya cordate total alkaloids auquired was 9.1%.2,A method for purification the Sanguinarine and chelerythrineA simple,convenient and rapid method for purification the sanguinarine, Chelerythrine was established,to use this method,the 87%sanguinarine,chelerythrine Can be easily obtained.3,The studied on the antibacterial effects of the macleaya cordate total alkaloids,total alkaloids chloride,Sangulnarine and chelerythrine to Phytophthora parasitica var.nicotianae and Pseudomonas solanacearum E.F.SmithThe results show that:the macleaya cordate total alkaloids had good antibacterial effects on Phytophthora parasitica var.nicotianae,the inhibition effects of 320μg/mL was 77.42%and EC₅₀ was 21.38μg/mL after 5 days.The macleaya cordate total alkaloids chloride also had good antibacterial effects on Phytophthora parasitica var.nicotianae,the inhibition of 320μg/mL was 86.54%and EC₅₀ was 16.22μg/mL after 5 days.The Sanguinarine had normal antibacterial effects on Phytophthora parasitica var.nicotianae,the inhibition effects of 40μg/mL was 62.54%and EC₅₀ was 21.38μg/mL after 5 days.The chelerythrine had weak antibacterial effects on Phytophthora parasitica var.nicotianae,the inhibition of 40μg/mL was 32.6%after 5 days.The macleaya cordate total alkaloids,total alkaloids chloride,Sanguinarine and chelerythrine had no antibacterial effects.4,The studid on the toxicity effects of the macleaya cordate total alkaloids,total alkaloids chloride to tobacco aphid and bean aphidThe macleaya cordate total alkaloids and total alkaloids chloride had good Toxicity effects on tobacco aphid and bean aphid.For tobacco aphid,this two reagents' LC₅₀ were 274.5μg/mL,501.19μg/mL after 60 hours;For bean aphid,this two reagents'LC₅₀ were 562.3μg/mL,380.2μg/mL after 48 hours or 30 hours.5,The studid on the Toxicity effects of the macleaya cordate total alkaloids,total alkaloids chloride,Sanguinarine and chelerythrine to Drosophila melanogasterThe macleaya cordate total alkaloids had good toxicity effects on Drosophila melanogaster,the LC₅₀ was 5.68mg/mL after 10 min.The macleaya cordate total alkaloids chloride also good toxicity effects on Drosophila melanogaster,the LC₅₀ was 6.5mg/mL after 10 min,Sanguinarine had none toxicity effects on Drosophila melanogaster,High concentration chelerythrine had toxicity effects to Drosophila melanogaster but need long time,light concentration had weak toxicity effects to Drosophila melanogaster. 6,The inhibition effects of the macleaya cordate total alkaloids to PVY by the field expermentThe macleaya cordate total alkaloids had a certain inhibition effects to PVY,by use the macleaya cordate total alkaloids,the PVY pathogen invading tobacco had been controled,but it was invalid if only one appliance the macleaya cordate total alkaloids,it need to appliance 2-3 times,then PVY pathogen invaded and pathogenesis level can be controlled.As the macleaya cordate total alkaloids consistency increasing, the inhibition effects to PVY also raising,light concentration had not obvious inhibition effects to PVY.
