In order to study the effect of dietary on the bacteria in the rumen of cows and mineral metabolism in the serum, and selecting strains of probiotics which were suit for the dietary of higher gradient. nine different diets (30:70, 35:65, 40:60, 45:55, 50:50, 55:45, 60:40, 70:30, 100:0) were used to fed Holstein cows during the dry period, Partial aerobe and facultative anaerobe were isolated, PCR-16S rDNA analyzed and detected. At the same time, Partial macro-element and microelement in the serum were detected. The result is reported as follows:1 56 strains of bacteria were isolated from the rumen fluid by artificial medium, and the 16S rDNA genes of 7 strains of representative bacteria were amplified by PCR. These bacteria were S.bovis(A0625, A1212, 0131), B.cereus(0113), B.licheniformis(0091), B.pumilus(0083) and S.thermophilus(0123) respectively by sequence homogeneous analysis. At the same time, the quantities of S.bovis, Bacillus and S.thermophilus were increased with the increase of gradients. S.bovis have remarkable difference (P<0.05) in 50-100% group and not remarkable difference (P>0.05) in 30-45% group; S.thermophilus have remarkable difference (P<0.05) in 30-55% group and not remarkable difference (P>0.05) in 55-70% group, and has remarkable difference (P<0.05) in 100% group compared with other groups; Bacillus have not remarkable difference (P>0.05) in 30%, 35% and 40%, and remarkable difference(P<0.05) in 45-100% groups. The results demonstrate that the dietary of different gradients has different effects on the quantity of aerobe and facultative anaerobe in the rumen, and the quantities of S.bovis was affected significantly. New evolutive branch of bacteria was not shaped by the phylogenetic analysis, however, there were several mutations in the nucleotide sequences of the S.bovis and S.thermophilus. 2 The concentration of serum Zn, Cu, Fe Ca, Mg increased gradually after the beginning of the treatment, reached the peak on 55% group and then decreased gradually, serum Cu, Fe, Ca, Mg have remarkable difference (P<0.05) in 55% group compared with 30%, 30% group(P<0.05)and no difference compared with 60%, 70%, 100%group. The content of Zn in serum have remarkable difference (P<0.05) in 50% group compared with other groups except 55% group. The concentration of serum P, Mn and K decreased gradually after the beginning of the treatment, respectively reached the peak on 50% and 55% group and then increased gradually, serum Mn have remarkable difference (P<0.05) in 55% group compared with 30%, 35%, 40%, 60%, 70%,100% group, serum K have remarkable difference (P<0.05) in 55% group compared with 30%, 35%, 40% group, serum P have remarkable difference (P<0.05) in 100% group compared with other groups, serum Na show rising trend and 55%, 60%, 70%, 100% group have remarkable difference (P<0.05) compared with 30%, 35%, 40%, 45%. The results demonstrate that the dietary of different gradients have different effects on the concentration of minerals in the serum of cows, the concentration of Ca, Mg, and K of the macro-element and Mn, Zn of the microelement were effected significantly.3 B.cereus, B.licheniformis and B.pumilus were isolated from the rumen of cows, who were fed by nine different diets. and these bacteria were suit for strains of probiotics for cows.
