Objective:To study the genetic diversity with ISSR-DNA and the HPLC fingerprint, effective components of Jizhao strain of Lonicera japonica Thunb. and explore their correclation. The data could provide conference evidence for dividing the cultivars resource and standardized planting of Lonicera japonica Thunb.Methods:The12Jizhao strain of Lonicera japonica Thunb. cultivars were analyzed by means of inter-simple sequence repeat (1SSR) markers. NTsys2.-10e software was used to process the data. The genetic similarity coefficients were calculated by the software. And the cluster analysis indicated the abundant genetic diversity in12Jizhao strain of Lonicera japonica Thunb. was constructed by using UPGMA method. The HPLC method was used to establish HPLC fingerprint and to determine the contents of chlorogenic acid, caffeic acid, luteolin and ischlorogenic acid A of Jizhao strain of Lonicera japonica Thunb.Results:15highly stable and repeatable ISSR primers (selected from50primers tested) detected a total of309bands, in which there were240polymorphic bands, and the percentage of polymorphic bands was77.67%. The result indicated that the Jizhao strain of Lonicera japonica Thunb. had abundant genetic diversity. The genetic similarity coefficients among12cultivars were from0.6408to0.8738. And the strain was divided into two clusters when similarity coefficient was0.68, Shandong and Henan Xinmi, which were correspondent with their geographical regions. HPLC method was used to build the chemical fingerprint of the12cultivars, definited13peaks in common, and the contents of chlorogenic acid, caffeic acid, luteolin and ischlorogenic acid A were all different.Conclusion:Through the research on ISSR genetic diversity and HPLC fingerprint, effective components of Lonicera japonica Thunb., found the cultivars had obvious regional characteristics. Clustering results from ISSR genetic diversity and HPLC fingerprinting were reflected the result. This indicated that geographical factors had a significant effect on Jizhao strain of Lonicera japonica Thunb..
