Effects On Photoqutotrophic Culture And Light-Emitting Diode(LED) In Vitro Paeonia Suffruticosa Shoots' Growth

This experiment used variety thee peony axillary bud as experimental material , used CP ,which is a kind of new cultivation vessel made of resin membrane material with higher permeability and lower moisture permeability as container, researching the variety tree peony plantlet cultivation at conventional cultivation conditions ,at applying co2 cultivation conditions and at allying new cultivation light source conditions (LED). The results are as follows:1. Using resin membrane made cultivation vessel (CP) to cultivate tree peony plantlet , the results indicated that: (1) Under normal culture,the plantlets growth of Paeonia'wulongpengsheng'grown in the culture pack system(CP) was better than the 100ml containers, while the plantlets growth of Paeonia suffruticosa'luoyanghong'and'huhong'grown in the two containers of various indicators of the difference was not significant (2)In point of using CP as cultivation vessel ,allying 3000ppm co2 can evidently promote the development of the plantlet and its quality, compared without the application of CO2 .2. Using CP as cultivation vessel and applying CO2 ,to compare the effect on the development of plantlet and its stomatal characters produced by different light intensity (1000Lx,2000Lx,3000Lx,)and sugar concentration(0%,1%,2%,3%).The results indicated that: (1) Improving the CO2 concentration in the CP and the light intensity can obviously increased the development of the Paeonia suffruticosa'wu long peng sheng'plantlet and the stomatal density of its leaves, it also can improved the development of Paeonia suffruticosa'hu hong'plantlet and it quality to some extent . But increasing the light intensity didn't obviously improve the quality of the Paeonia suffruticosa'luo yang hong'plantlet , the treatment using 3000Lx and 2000Lx has no significant difference .(2)In the case of applying CO2 , variety of tree peony plantlet can grow normally without the application of sugar in the cultivation media by photoautrophic mode, but the growth velocity is slower and the plantlet is weaker. The plantlet of Paeonia suffruticosa'wu long peng sheng'and'luo yang hong'growed well in the media with a sugar concentration of 2%, but a 2% concentration of sugar can satisfy the growth of Paeonia suffruticosa'hu hong'plantlet. It showed that when applying CO2 on variety tree peony plantlet, the sugar added in the media is necessary, but it can be reduced by somewhat.3. Using controllable Light quality ratio and Optical quantum number red and blue LED light source as cultivation light source, the plantlet was cultivated with the application of CO2.The result showed that: the growth index of Paeonia suffruticosa'wu long peng sheng'and'luo yang hong'plantlet changed in a relativity uniform trend. The treatment effect was much better in point of a light source of 75%R and 25%B, while the Paeonia suffruticosa'hu hong'plantlet got a better effect at a light source of 50%R and 50%B.But at a light source of 100%R and 100%B the plantlet grown lower and weaker, and compared with the other three kinds of light treatment, there was a significant difference.4. In the case of applying CO2 to CP vessel and the LED light source was 75%R and 25%B,using the best light intensity and sugar concentration for the growth of Paeonia suffruticosa'wu long peng sheng'plantlet ,to cultivate the root-induced Paeonia suffruticosa'wu long peng sheng'plantlet , the results showed that:compared with the normal condition, while the plantlet was cultivated under a regulator environment , its leaf length,leaf amplitude,plant height,chlorophyll index improved obviously, the quality of root-induced Paeonia suffruticosa'wu long peng sheng'plantlet was also improved evidently.This research applies the CO2 enrichment and the light-emitting diode into the culture of the Paeonia suffruticosa in vitro, discussed the series of changes about the form indicators of Paeonia suffruticosa'wulongpengsheng',Paeonia suffruticosa'luoyanghong'and Paeonia suffruticosa'huhong'under the different environmental conditions.Can improve the quality of the plantlets of Paeonia suffruticosa'wulong pengsheng'.
