In order to commercially and industrially obtain high quality in vitro shoots, this thesis uses Chrysanthemum morifolium 'A9(14)', 'donglinruixue' and 'xiangxue' as experimental model plants which cultured by some new technologies including Light Emitting Diode (LED) , carbon dioxide, novel enveloped-shaped vessels made of a fluorocarbon polymer film which possesses gas permeability, light transmittance, thermal stability and heat seal process ability and rock wool system. The main results obtained are as below:1. Practical application of Light Emitting Diode(LED)in in vitro Chrysanthemum morifolium shoots. Compared to fluorometry, LEDs accelerate development of in vitro Chrysanthemum morifolium shoots more effectively. In this thesis, the best red/blue light ratio for development of in vitro Chrysanthemum morifolium shoots is 7:3. Under red/blue light ratio 7:3, the best irradiation is 60μmol·m-2·s-1 .2. Practical application of vessels made of a fluorocarbon polymer film and rock wool system in in vitro Chrysanthemum morifolium shoots. Under normal and sugar-free culture condition, the Chrysanthemum morifolium shoots develop better in vessels made of a fluorocarbon polymer film than in glass bottles. At the same time, the study substantiates that rock wool system is better than agar sustain culture medium.3. Culture in vitro Chrysanthemum morifolium shoots with sugar-free culture medium in CO2 enriched condition. CO2 could replace sugar in in vitro Chrysanthemum morifolium shoots culture. It has 1500ppm, 3000ppm and 4500ppm three CO2 concentration in this thesis. The best CO2 concentration for development of in vitro Chrysanthemum morifolium shoots is 3000ppm. The development of little shoots in CO2 enriched condition at 1500ppm is close to that of in normal condition with 3% sugar in culture medium.The above recoveries supply the theoretical and practical evidences for effectiveness and culture conditions of the new technology in in vitro Chrysanthemum morifolium shoots.
