| Starch is one of the most important raw materials for food and industrial applications. High amylose maize kernel has become the main source of industrial raw material. Study on the interaction between ae and other mutant gene of amylose and also study on the effect on ae expression of different genetic backgrounds, and then screen out the genetic materials which make ae expressed efficiently to provide new high amylose maize germplasm resource.Four high amylose maize inbred lines and six normal inbred lines were used as materials.The differences among sequence of ae gene of different maize were investigated, which established a good base to mark gene ae and shorten the period of breeding. The two representative varieties of high amylose,six common maize inbred lines and their hybrids were also determined of the content of amylose, weight of 1000-grain, activity ofα-amylase, germination percentage and germination potential. The main results as follows:1. According to the sequence of gene ae reported in Genbank, 20 pairs specific primers were designed and carried out PCR above materials. PCR production were separated and the sequence were measured.The results indicated that the sequence of gene ae selected in this research was quite conservative,and only one deletion with seven bases(CGGCAAG)at locus 9658 was found;one insert with one bases at locus 10144,two transversions with one base(T→C) at locus 10390 and(C→T) at locus 10430.2. The results showed the content of amylose and weight of 1000-grain that controlled by gene ae have obviously hybrid vigor. It is concluded that the amylose content in the kernels can be remarkably improved when they are introduced with gene ae by backcross and the main influence of genetic background on the amylose content is non-additive effect. It may be effective to improve to gather modificatory genes by recurrent selection.The activity ofα-amylase at 48hours, percentage and germination potential were also tested. It is found that the activity ofα-amylase at 48hours, germination percentage and germination potential of the hybrid is higher than ae high-amylose and inbred lines obviously ,and high-amylose maize are lower than commom inbred lines.Breeding of high-amylose maize should not only paying attention to improving the amylose content,but also paying attention to selection of kernel quality. |
