Tring to culture BHK-21 cell line with low serum concentration and further to produce foot-and-mouth disease vaccine, in this study BHK-21 cell line was first adapted to culture medium with low serum concentration, and then cell growth situation and rivus titre were compared under different serum level.The result showed that growth rate of BHK-21 cell line adapted to MEM SLM and cell number was similar with control group, so BHK-21 cell seed banks with low serum culture medium were established. Compared with cell number, growth rate, cell activity, virus titer, production cost and production efficiency under different serum concentration, 3% serum concentration applicable to actual production was defined. At this level it indicated that cell number, growth rate, cell activity, virus TCID50 and LD50 were①2.288×107, 2.03, 95.3%, 7.83 and 8.00 in small square bottle(T75) respectively;②5.871×108, 1.70, 94.6%, 7.67 and 7.89 in 3000ml roller bottle respectively and③7.586×109, 1.73, 95.0%, 7.72 and 8.22 in 15000ml roller bottle respectively. So it is potential to replace traditional culture medium with low serum culture medium MEM SLM in FMD vaccine production.
