In this study, core primers were defined from preliminary screening by fluorescent SSR marker technique to study genetic diversity of 194 skeleton and innovation maize inbred lines in Jilin province. At the same time, constructed morphology and fingerprint database of maize inbred lines. The results were summarized as follows:1. Defined 25 pairs of SSR core primers with sharply peak value and high polymorphism from 72 pairs of SSR primers with average distribution, clear band patterns, good reproducibility and less date missing by fluorescent Capillary Electrophoresis. Defined preliminary 12 pairs core primers based on amplified bands definition, reproducibility, PIC value, fragments size and parents complementary bands pattern to construct the fingerprint database of maize inbred lines.2. Studied the genetic diversity of 194 maize inbred lines by fluorescent SSR marker technique, detected total 266 alleles, the average alleles per locus were 10.6 with average from 3 to 23. Alleles were from 4 to 15 of 86% SSR locus. The average PIC value per SSR locus was 0.657 ranging from 0.301 to 0.899. The most alleles were up to 23 and the most PIC value was 0.899 detected by bnlgl61k8. The least alleles were 3 detected by phi072k15 and bnlgl007y5; meanwhile, bnlgl007y5 had the least PIC value which was 0.301.3. 194 inbred lines were divided into 8 genetic groups by cluster analysis, which were six groups including Zi330, Lvdahonggu, Mo17 PB, Sipingtou, improved Reid and the other two groups. Inbred lines of which belonged to Zi330 group occupied 9.8 percent, belonged to Mo17 group occupied 13.9 percent, belonged to Lvdahonggu group occupied 11.9 percent, belonged to PB group occupied 7.7 percent, belonged to Sipingtou group occupied 6.7, belonged to improved Reid group occupied 15.5 percent; in the other two groups, belonged to foreign groups occupied 20 percent more than 6.7 percent of the other one. The genetic composition and the pedigree of some known inbred lines were analyzed. The result of clustering by SSR were well consistent with family tree.4. Constructed fingerprint of maize inbred lines based on 12 pairs of fluorescent core primers, and preliminary morphology and fingerprint based on morphology traits, which can identify maize inbred lines easily and accurately.
