The main crops technical regulations of varieties dentification have been promulgated using SSR molecular markers in China,such as: Maize(NY/T1432-2014),Wheat(NY/T2470-2913),Rice(NY/T1433-2014),Sorghum(NY/T2467-2013),Sybean(NY/T2595-2014),etc.The construction of DNA fingerprint database based on crop industry standards is also carried out.On the basis of the maize ndustry standard,the maize standard DNA fingerprint database was established,and a mature system was established,which provided a reference for other crops.The characteristics of crops directly affect the construction of different programs,the degree of difficulty is not the same.In order to play the role of the main crop variety DNA fingerprint database in the identification of crop varieties as soon as possible,we need to find a general,simple and convenient database construction scheme.In this study,4 kinds of main crops of maize,sorghum,soybean and wheat were studied to establisha general DNA fingerprint databas construction scheme.1.Take 166 sorghum varieties and the standard varieties of the 6 maize as an example,in comparison there are two kinds of crop establishment scheme,and through the two crop establishment scheme proved: construction method of maize standard DNA fingerprint database showed more stable,simple,accurate,and fully apply the sorghum scheme.Therefore,on the basis of the corn standard,166 sorghum varieties DNA fingerprint database were established,and the suspected varieties were screened,and the list of suspected samples was determined.Through one year field test results show that the identification results of field and laboratory test results are basically the same phenotype,further explains the corn standard DNA fingerprint database construction scheme can be applied in the DNA fingerprint database of cross sorghum in the construction,but also laid the foundation for the exploration of general construction of other crops couffand case.2.Take the 5 soybean varieties as an example,to explore the corn DNA fingerprint library construction in Soybean.Through the comparative analysis of the whole experimental process,it was found that soybean could be applied to the construction of corn standard DNA fingerprint database.In the process of theestablishment of universal building program,the main problem is a processing method of sample problems,followed by the primer annealing temperature is not uniform,the establishment of PCR reaction system and reaction procedure is relatively difficult.But for the other ones can link and corn standard DNA fingerprints on the general scheme,such as the data analysis because soybean belongs to the diploid self pollinated crops,with relatively simple,in the corn data to be analysed parameters of the effect is good,easy to automate the analysis of data.3.Take the 5 wheat as an example,to explore the corn standard DNA fingerprint database construction in Wheat.Through the contrast analysis of the whole test process,the basic can be constructed for wheat maize standard DNA fingerprint database.The main problems existing in the construction is the universal primer,six times the body belongs to wheat heterologous,peak type itself is more complex,in the areas of data analysis using data mining analysis parameters of diploid,interference peak more serious,need a lot of manual correction,automatic data analysis more difficult.Through the 4 representative crop: diploid cross-pollination of corn,diploid often cross-pollinated sorghum,diploid self-pollination of soybean,allohexaploid wheat,explore the universal building program and summed up the limiting factors to realize the general construction scheme,and proposes solutions to these restrictive factors,as to provide the reference for each crop industry standard developed and optimized,lay the foundation for the aim of realizing the unified database scheme of different crops standard.
