Study On Identification Of The Purity Of Hybridization Rape By Polyacrylamide Gel Electrophoresis

On the basis of research on seed purity identification,analyzes the influence of scanning electron microscope,esterase isozyme,mixed solution of cationic polyacrylamide and SDS,acetic acid-page,lactic acid-page,formic acid-page on hybridization rape seed.Verification test is carried on.The results are as follows.1.Analyzes esterase isozyme in different hybridization rape varities.Choose zero day,fist day,third day,fifth day and seventh day germination developmental stages to electrophoretic.The result showed that,the third and fifth day buds and seedlings are the most appropriate identification period for variety identification authenticity purity.2.Research on the basic electrophoresis buffer system in the varieties of hybrid rape seed purity and authenticity of identification.Rape of albumin,globulin,glutenin and gliadin proteins such as the four SDS-PAGE patterns,and explore suitable varieties of hybrid rape identification of protein electrophoresis system and found that separation of plastic,rubber enrichment concentration,ion strength,electrode buffer concentration can affect the electrophoresis results.The analysis,the choice of trial separation of plastic 10%,3% concentration of plastic,alkali electrode buffer PH8.3,acid electrode buffer pH3.4 a combination of protein electrophoresis,the patterns in a clear,better-performing.The application of the method of eight hybrid rape combination of dry seeds were analyzed. The results showed that the four main proteins in the inter-species there are different degrees of difference in the band,can better distinguish between different varieties of hybrid rape.One polymorphism high-protein,high resolution map,the identification of hybrid rape seed varieties authenticity and purity better.3.Analysis of the acetic acid - PAGE,lactic acid - PAGE,formic acid - PAGE three acid electrophoresis buffer system in the rape-protein,globulin,glutenin and gliadin proteins in four categories,such as hybrid rape seed varieties and authenticity Identification of purity. The results showed that although the technical stability,rapid,and the economy,the acid electrophoresis system of spectrum than obvious effect of the basic system of protein electrophoresis.Most acid electrophoresis buffer system of the spectrum less effective,but in acetic acid - PAGE buffer system conditions,the strong-protein polymorphisms, identified species of hybrid rape relatively good results.4.To explore the interior of SDS-PAGE analysis and field testing and purity of the relationship between the choice of eight varieties of hybrid rape,through the cultivation of field and laboratory tests of purity of hybrid variety of protein-SDS-PAGE of detection verification test results Correlation Analysis.The results showed that the purity and protein-seizure spectrum purity was very significant positive correlation r=0.8587 (r~2=0.7373),a field-protein purity by SDS-PAGE of the purity of the bands regression equation,Y=0.6988X+28.992.
