Dendrobium huoshanese CZ Tang et S.j.cheng is treasure in medicinal Dendrobium,produced in Huoshan County,Anhui Province.The growth of it demands extremely harsh conditions,and seed multiplication capacity is very low.due to excessive exploitation,natural resources have been tialed off,it is difficult to carry out large-scale artificial cultivation for the transplant of test- tube plantlets of D.huoshanese have a lower survival rate.At present,there are have more of report in artificial seeds,but artificial seeds of D.huoshanese have not been reported,therefore,we can consider the adoption of rapid propagation of artificial seeds as a effective way to solving lack of resources.In this paper,the artificial seeds of D.huoshanese was produced with Auxiliary buds, Protocorm-like bodies,and adventitious shoots.By using of single-factor and orthogonal experiment,studied on the effect of 5 factors(Maltose(%),hormone rate between 6-BA (mg·L-1) and NAA(mg·L-1),active carbon(%),sodium alginate(%),time of ion exchange(min)) on germination rate of artificial seeds of D.huoshanese,the effect of the type and size of propagators on germination rate and rate of seedlings.Then the leaking rate of maltose and variation of pH value of artificial seed capsule during vegetating of artificial seeds of D.huoshanese was measured,also we studied on the seedling plants growth characteristics,artificial seed storage,preservation,and other aspects in order to provide technical methods for depth study to artificial seeds D.huoshanese.The main results of study were followed:1 From single-factor test that,the maltose is suitable carbon,appropriate concentration is 4.0%,by orthogonal design testing,Maltose plays the most important role in inducing the D.huoshanese artificial seeds to germinate,the most appropriate combination is: maltose4.0%,hormone rate between 6-BA(mg·L-1) and NAA(mg·L-1)10:1,active carbon0.1%,sodium Alginate 4%,time of ion exchange is 10 min.Embedded in this formula,the germination rate of artificial seeds of D.huoshanese takeing auxiliary buds, protocorm-like bodies,and adventitious shoots as the propagators is 65.6%,90.1%, 75.2%.After germination,the survival rate of seedlings of artificial seeds was 80.6%, after germination,the survival rate of seedlings of artificial seeds was 16.1%,80.6%, 19.1%. 2 The appropriate size of auxiliary buds and adventitious shoots using as propagators 0.6-0.8 cm,the diameter of protocorm-like bodies is 0.3 cm,protocorm-like body is more suitable for embedding.3 The leaking rate of maltose of artificial seed capsule was 0.52%,it is result of requiring a higher concentration of maltose,the pH value of artificial seed capsule decreased during the process of vegetation of artificial seeds,the pH value of artificial seed capsule using auxiliary buds,protocorm-like bodies,and adventitious shoots as the propagators dropped from 5.60 to 4.69,from 5.60 to 4.80,from 5.60 to 4.71,the pH values gradually Reducing may impede artificial seeds into seedlings.In addition,the comparison plants growth characteristics is better than artificial seeds seedling plants,artificial seeds capsule can not fully meet the the nutrients of propagators requiring therefore,the quality of seeds need to be improved.4 The plants growth characteristics of artificial seeds using protocorm-like bodiesas the propagators is better than artificial seeds using auxiliary buds,and adventitious shoots, the plants height of it was 2.36 cm,stem diameter is 1.25 mm,fresh weight is 186.23 mg, dry weight is 15.01 mg,the chlorophyll content is 0.31 mg/g·FW.5 After having been stored at 4℃for 20 d,the germination rate of artificial seeds of D.huoshanese takeing Auxiliary buds,Protocorm-like bodies,Adventitious shoots as the propagartors are 3.3%,10.6%,5.2%.And in natural conditions the germination rate is 13.8%after 10.0 g/L carbendazim haven been appended into artificial seed capsule,this result would be a foundation of manufacture and further study of the artificial seeds of D. huoshanese.
