Objective:1.To study the dynamic accumulation of chemical components of Dendrobium huoshanense under different cultivation modes,and explore the impact of different cultivation modes on the accumulation of active components of Dendrobium huoshanense.2.To study the correlation between the matrix components and the accumulation of active components of Dendrobium huoshanense,and to further understand the differences in the planting modes of Dendrobium huoshanense,the differences in the matrix of different planting modes were studied.3.To study the effect of climate factors on the accumulation of effective substances in Dendrobium huoshanense,and to explore the changes in the content of effective substances and growth indicators of Dendrobium huoshanense under different climate factors.Method:1.Polysaccharide extraction and content determination and polyphenol extraction and content determination of Dendrobium huoshanense stem from different cultivation modes for 1-3 years were carried out to compare the content accumulation differences between different cultivation modes,the crude polysaccharide extracted was refined and hydrolyzed to explore the monosaccharide composition differences of Dendrobium huoshanense polysaccharides from different cultivation modes for 1-3 years,and the ethanol extract of Dendrobium huoshanense stem was analyzed by GC-MS,To explore the differences of chemical components of ethanol extracts from Dendrobium huoshanense stem in different cultivation modes for 1-3 years.2.Determine the total phosphorus content,total nitrogen content,nitrate nitrogen content,ammonium nitrogen content and pH value of different cultivation modes of Dendrobium huoshanense,and compare the physical and chemical properties of different cultivation modes.Determine the trace elements of different cultivation modes by ICP-MS,and further compare the differences between different cultivation modes.3.The effects of different climatic factors on the quality of Dendrobium huoshanense were compared through the determination of polyphenol content,polysaccharide content and growth index of Dendrobium huoshanense after the treatment of different climatic factors.Result:1.Polyphenols of Dendrobium huoshanense under different cultivation modes accumulated to the highest level in the second year,and then decreased.The polysaccharide content of Dendrobium huoshanense cultivated on coconut shell substrate is the lowest,and that of Dendrobium huoshanense cultivated on bark substrate is the highest.The polysaccharides of Dendrobium huoshanense in different cultivation modes decreased with the increase of years,with the highest average content of about 12%in the first year;The lowest average content is about 6%over the third year.The polysaccharide content of Dendrobium huoshanense cultivated on coconut shell substrate is the lowest,and that of Dendrobium huoshanense cultivated on bark substrate is the highest.The polysaccharides of Dendrobium huoshanense in different cultivation modes decreased with the increase of years,with the highest average content of about 12%in the first year;The lowest average content is about 6%over the third year.The polysaccharide composition of Dendrobium huoshanense is mainly glucose,mannose,galactose,xylose,arabinose and rhamnose.The cultivation period and cultivation mode have no effect on the monosaccharide composition of Dendrobium huoshanense,but have a significant impact on the monosaccharide proportion of the polysaccharides.2.The total phosphorus content of coconut shell>pear>stone,the total phosphorus content of bark matrix and pear matrix is close,and the total phosphorus content of coconut shell matrix is 86.2 mg/kg,which is much higher than that of other matrices.The results of the determination of total nitrogen content in the matrix showed that the stone matrix did not contain nitrogen,and the total nitrogen content in the coconut shell matrix was about three times that in the pear tree and bark matrix,and the nitrogen content in the pear tree and bark matrix was similar.Nitrate nitrogen content and ammonium nitrogen content,nitrate nitrogen and ammonium nitrogen content results are similar to total nitrogen content results.The total nitrogen content in coconut shell matrix is about three times that in pear and bark matrix,and the nitrogen content in pear and bark matrix is similar.In terms of trace elements,there is a big difference between the stone cultivation substrate under the forest and the other three cultivation substrates.The composition and content of metal elements in pear matrix and bark matrix are the closest.3.Temperature,humidity and light intensity have a certain impact on the accumulation of polyphenols in Dendrobium huoshanense.In general,temperature and humidity have the greatest impact on the accumulation of polyphenols in Dendrobium huoshanense,while light has a small impact on it.Temperature has a great impact on the accumulation of polysaccharides in Dendrobium huoshanense,followed by humidity.In the light intensity range of 2000 lux-12000 lux,about 6000 lux is the most suiTab.le for the accumulation of polysaccharides in Dendrobium huoshanense.Temperature has the greatest influence on the growth of Dendrobium huoshanense.When the temperature is 35℃,the content of polysaccharides,polyphenols,the number of new stems,the increase of stems and the drying rate are the lowest.The higher the humidity,the better the growth of Dendrobium huoshanense.
