The text had carried through not only the systematic research on the rapid propagation of Dendrobium huoshanense,but also the analytical research on the regulatory of secondary metabolism in Dendrobium huoshanense with the following main results:1,The four factors (MS,6-BA,KT,NAA) were selected,and each factor had there levels.Through the orthogonal design, the tufted-bud induction of Dendrobium huoshanense was studied ,and the results showed that the orders of four factors to influence the tufted-bud induction of Dendrobium huoshanense were : KT> minimal medium >6-BA>NAA,of which it was very significant that KT was effected on the tufted-bud induction of Dendrobium huoshanense,but minimal medium,6-BA,NAA were not.The optimal Culture Media of he tufted-bud induction of Dendrobium huoshanense was screened out,it was that:1/2MS+0.2mg/LKT+0.2mg/LNAA+0.3mg/L6-BA2,Medium of 1/2MS+0.2mg/LKT+0.2mg/LNAA+0.3mg/L6-BA as control,we added separately mashed potato,banana mud,Raphanus Sativus of which chroma also was100g/L,200g/L,300g/L,and lanthanum nitrate of which chroma was 10mg/L,20mg/L,40mg/L to culture Dendrobium huoshanense.The results showed that the different additive had some promoting effects on growth of Dendrobium huoshane -nse,especially with mashed potato and lanthanum nitrate, it had remarkably discrepancy.At the same time,with correlation analysis on physiological index,con -tent of chlorophyll can be acted an important physiological index of early selection on qualitical nursery stock of Dendrobium huoshanense.3,Medium of MS as control, we added separately IBA(0.5mg/L,1.0mg/L,2.0mg/L ),NAA(0.5mg/L,1.0mg/L,2.0mg/L) and banana mud(10%,20%,40%) to use as the rootage culture.The results showed that when banana mud of 20% was added,it was the best effect on rooting of Dendrobium huoshanense.4,Medium of 1/2MS+0.2mg/LKT+0.2mg/LNAA+0.3mg/L6-BA as control, we added separately L-lys(0.5mg/L,2.5mg/L,12.5mg/L),sys(0.2mg/L,1.0mg/L,5.0mg/L) and lanthanum nitrate (10mg/L,20mg/L,40mg/L) to control research about polysaccharides in Dendrobium huoshanense.The results showed that L-lys(2.5mg/L) and lanthanum nitrate(20mg/L) could improve evidently polysaccharides in Dendrobium huoshanense within each definite concentration range,and it had remarkably discrepancy,but sys had only discrepancy.At the same time,we carried out preliminary study on which polysaccharides in Dendrobium huoshanense clean out DPPH of organic free-radicals,and found that polysacchari- des in Dendrobium huoshanense had the scavenging activities.5,Medium of 1/2MS+0.2mg/LKT+0.2mg/LNAA+0.3mg/L6-BA as control, we added separately L-lys(0.5mg/L,2.5mg/L,12.5mg/L),sys(0.2mg/L,1.0mg/L,5.0mg/L) and lanthanum nitrate (10mg/L,20mg/L,40mg/L) to control research about dendrobine and moscatilin of bibenzyls,usingHPLC/UV to determine conten -t of it.The results showed that L-Lys and sys had the promoting effects on content of moscatilin; L-lys(12.5mg/L) and sys(0.5mg/L) could improve greatly Moscatilin within each definite concentration range.
