Lanzhou lily is the variety of the david lily, perennial herb,with rich nutrition, has high nutritional quality, which has famous for nationwide by famous dish and good medicine.there were a little of report about the tissue culture of lanzhou lily.especially there was no roport about the embryo invitro culture selecting the scale, embryo,leaves as explants for tissue culturing .this research is divided into three phages: establishment of clone system,multiplication and differentiation ,root and bulblet formation.Optimizing the best sterilization method: washing by satrtated detergent-ddH2O-70%-anhydrous alcohol 1-2min-washing ddH2O-15min by 15% disinfection solution—5-6min by 0.1% hydrargyrum—washing 3 times by ddH2O. Sterilizationg the inner, middle and ourer level of flakes by the same sterilizationg method. It is different for layer to sterilizate.Stages of establishment of clone system:In the progress of primary culture of the Lilium davidii var. Unicolor, the culture medium which fit for primary culture of the flake of the Lilium davidii var. Unicolor was MS+1.0mg/L BA +0.2mg/L NAA. Treating with lower temperature to flake can influence the primary culture。As the explant, the differentiation rate of the squama of the bulb which has treated by lower temperature was much higher than un-treated with the lower temperature. The culture medium which fit for primary culture of the root of the Lilium davidii var. Unicolor was MS+BA2.0mg/L+NAA0.2 mg/L ,MS+BA2.0mg/L+NAA0.1 mg/L. The culture medium which fit for primary culture of the Leaves of the Lilium davidii var. Unicolor was MS+1.0mg/L BA +0.2 mg/L NAA,MS+1.0mg/L BA +0.1 mg/L NAA. In the culturing of young embryo, the appropriate culture medium which was used for culturing the lily embryo was not found. After biology statistic analysis, the young embryos were random arising, which did not have the connection with the culture medium. Treating with the lower temperature also can not influence bourgeoning the embryo.Stages of multiplication and differentiation:During the Stage of multiplication and differentiation, it is found that, the culture medium which fit for Stage of multiplication and differentiation of flake of the Lilium davidii var. Unicolor was MS+0.5 mg/L BA +0.5 mg/L NAA. The culture medium which fit for Stage of multiplication and differentiation of Leaves of the Lilium davidii var. Unicolor was MS+2.0 BA mg/L+0.4 mg/L NAA. The culture medium which fit for Stage of multiplication and differentiation of Leaves of the Lilium davidii var. Unicolor was MS+1.5mg/L BA+0.5 mg/L NAA. The culture medium which fit for Stage of multiplication and differentiation of embroy of the Lilium davidii var. Unicolor was MS+1.0 mg/L BA +0.1 mg/L NAA.Stages of root and bulblet formation:During the Stage of root and bulblet formation, the result showed that the culture medium which fit for Stage of root and bulblet formation of flake of the Lilium davidii var. Unicolor was 1/2 MS+0.2 mg/L NAA + surgar 60g/L. The culture medium which fit for Stage of root and bulblet formation of root of the Lilium davidii var. Unicolor was1/2 MS+ 0.3 mg/L NAA + surgar 60g/L. The culture medium which fit for Stage of root and bulblet formation of leave of the Lilium davidii var. Unicolor was1/2 MS+ 0.3 mg/L NAA +surgar60g/L.The effects of initial medium on explant differentiation: the differentiation of lower flakes was higher than that from other place. While the differentiation of the base of leaves was greater than that of the middle or the top of leaves. And the differentiation of the base of the root was significantly higher than that of the middle and the top of root.The effects of subculture medium on explant propagation: the proliferation of adventitious buds were the best.The buds from the base of root had higher proliferation multiple significantly than the middle and the top of root, as well as the buds from the base of leaves had higher proliferation multiple significantly than the middle and the top of leaves.The effects of rooting medium on explant induced rooting: the growth of individual root from the lower of flake was the best.
