BC2F11/BC2F12 derivatives of[T.aestivum c.v.Chinese spring(CS)×T.aestivum c.v Inayama Komugi- R.ciliaris amphiploid]were screened by a combination of multiple techniques,such as spike morphology observation,root tip cell(RTC) chromosome sequential C-banding and GISH,pollen mother cell at metaphaseⅠ(PMC MI) configuration analysis and fluorescence in situ hybridization(FISH).Ten disomic addition lines were identified.A total of 135 EST,27 STS and 253 SSR primer pairs located in 7 homoeologous groups of wheat and barly were used for amplification of 27 wheat-R,ciliaris disomic addition lines developed by Wang et al(1997,2000) and Li(2005) and the present research. Fifty-five primer pairs could amplify polymorphic bands between common wheat variety "Inayama Komugi" and the Inayama Komugi- R.ciliaris amphiploid,and 31 of the 55 could amplify the specific bands of R.ciliaris in the addition lines.According to PCR results,the added R.ciliaris chromosomes in lines 07K02,07K06,07K39,07K41,07K43,07K201, 07K202,07K255 and 07K256 belong to homoeologous group 1 of wheat.The added R. ciliaris chromosomes in lines 07K07,07K08,07K09,07K11,07K14 and 07K17 belong to homoeologous group 2 of wheat.The added R.ciliaris chromosomes in lines 07K15, 07K16,07K21,07K22 and 07K47 belong to homoeologous group 6 of wheat.In summarize,lines 07K02,07K06,07K39,07K41 and 07K43 had the same alien chromosomes;07K201 and 07K202 added the same chromosomes;07K255 and 07K256 added the same chromosomes;07K07,07K08,07K09 and 07K11 had the same alien chromosomes;07K15 and 07K16 added the same chromosomes;07K21 and 07K22 added the same chromosomes.Fourteen primer pairs identified could be used for tracing different R.ciliaris chromosomes in the wheat background,respectively.The identified disomic addition lines were further evaluated for wheat scab resistance. From three years's evaluation results,lines 07K39,07K41,07K43,07K201,07K202, 07K.255,07K256 showed similar resistance level as the resistance control,indicating they might be new genetic resources in wheat breeding for scab resistance.
