| Tissue culture is already a common procedure for plant rapid propagation and a simple and effective way to breed precious flowers varieties for virus-free plants could be obtained rapidly by tissue culture.The systemic research was carried out by establishment of rapid propagation sysytem,enlargement and redifferentiation of bulblets in vitro,physiological detection after transplantation on Lilium oriental 'Siberia"Sorbonne' and Lilium longiflorum 'White Heaven'.With the scales and bulblet leaves in vitro as explants and by inducing adventitious buds directly,factors related to micropropagation process including culture initiation,proliferation, roots induction and transplantation were examined highly detailed.1.Regeneration frequency could be more than 80%in MS base medium supplemented with appropriate concentration of NAA and 6-BA which was efficient for inducing adventitious buds.2.Mass shoot regeneration occurred from the base part of bulbiet leaves.The 'siberia"White Heaven' had stronger ability to induce adventitious buds than 'Sorbonne'.Regeneration frequency could be more than 86%in the medium supplemented with NAA/IBA and 6-BA on which better bulblets were observwd.The best phytohormone combination were NAA 0.2mg/L+KT 0.4mg/L,NAA 0.1mg/L+KT 0.2mg/L,IBA 0.1mg/L+KT 0.2mg/L for 'White Heaven' 'Siberia' 'Sorbonne',respectively.3.The roots could regenerate in the proliferation medium while the bulblets in vitro had strong ability to rootage.4.Surivival rate of plantlets could reach more than 95%with bulblets diameter≥1 cm when transplanted to pearlite/soil(3:1).The experiment about actors affecting rapid propagation system of bulblets in vitro was carried out by MS medium supplemented with accretions.The best mediums for 'sorbonne' and 'siberia' were MS medium suppletmented with 30g/L sucrose and 3mg/L Paclobutrazol, respectively.Optimum bulblets enlargement and redifferentiation of 'white heaven' were observed in MS medium with 60g/L sucrose and 3mg/L Paclobutrazol.The diameter of bulblets and propagation rate of Lilium oriental could reach above lcm and 8 respectively in the mediums mentioned above.The better growth of bulblets in vitro was observed in MS medium with 4~10g/L and 10g/L AC for 'White Heaven' 'Sorbonne' and 'Siberia'.The healthy plantlets surviving from transplantation were foliar application with Paclobutrazol after transplanting to soil.The leaves photosynthesis was improved, chlorophyll a+b content,soluble protein and sugar content enhanced while water content decreaced with 500mg/L treatment.The growth and development was improved after the transplantation to soil under treatment. |
