OBJECTIVE: To examine the effects of jaagsiekte sheep retrovirus(JSRV-NM) on the clinical pathological manifestation, the changes of lymphocytes subsets, lymphocytes apoptosis, blood-gas of sheep experimentally infected with JSRV-NM strain. METHODS: In March 2008, five newborn lambs within 24 hours after their birth were inoculated intratracheally with cell-free filtration of pulmonary tumour tissue containing JSRV-NM strain from natural cases, 1.0ml per lamb. At different post-infection, vein blood samples were collected, and the lymphocytes apoptosis number, CD4+T and CD8+T lymphocytes were examined by using the flow cytomytor. At the same time, the pH,pCO2,pO2 BE(Base excess,), BB(Buffer base),HCO3std ,HCO3act, TCO2(Total carbon dioxide), cH+(hydrogen ion concentration,), O2SAT(O2 saturation,), O2Cont(Oxygen content), ctHb(est),AnGap, K+(Kalium ion concentration), Na+(Sodium ion concentration), Cl-(Chlorine ion concentration,),and the activity of CK, LDH, ALP, ALT, AST were inspected. RESULTS: The experimental group presented some clinical signs of the sheep pulmonary adenomatosis(SPA) . The body temperature was normal, the pulse rate and the breathe rate higher than the normal range. At 30 d post-infection, the relative percentage of CD4+T lymphocytes and the relative percentage of CD8+T lymphocytes were higher then the control group, and decrease subsequently, and then show a fluctuation. At 30 d post-infection, the early and late lymphocytes apoptosis number lower than the control group, undulating in the after time. In different time periods,AnGap,pO2,HCO3act,HCO3std,ctCO2,O2SAT,O2CT,Hct,ctHb(est) of analysis of blood-gas and ALT,AST,LDH , CK , ALP of serum enzymes showed significant difference between the experimental group and the control group. CONCLUSIONS: The typical symptoms of SPA has not appeared, and the disease was in the initial stage. The changes of CD4+T, CD8+T lymphocytes in peripheral blood of sheep experimentally infected with JSRV-NM strain demonstrated which may be the result of cell immunological reaction and the lymphocytes injury. Periphery lymphocyte apoptosis in sheep experimentally infected with JSRV-NM strain was found,lymphocyte apoptosis may be one of the most important clinical symptoms. The detection of blood-gas and the serum enzymes of sheep experimentally infected with JSRV-NM strain was helpful for diagnosis of SPA.
