Studies On Association Of Fatty Acid Desaturase Gene Polymorphism With Some Fatty Traits In Three Chicken Breeds

The research adopted the molecular quantitative genetics method,used gas chromatography-mass spectrometry technology and Mr.Suo's distill method to mensurate the fatty traits and PCR-SSCP method to detected the fatty acid desaturase gene polymorphism. To studied on the characteristics of deposition in different breeds,different periods and different sex and the possibility to used the△6,△9 fatty acid desaturase gene as the molecule marker. The results were as follows:1.Determined the fatty acids contents in breast and leg muscle in the three chicken groups.The difference of breast fatty acids contents was significantly in different periods(P<0.05).The polyunsaturated fatty acids(PUFA) and essential fatty acids(EFA) contents were highest on 4 week and was decreased in 5 periods. The unsaturated fatty acids(UFA) wasn't significantly defference in different periods(P>0.05).The most leg fatty acids contents wasn't significantly difference in the different periods(P>0.05). Mostly, The difference of seven saturated fatty acids(SFA) contents was significantly in 4 periods(P<0.05) and highest on 4 week.The PUFA and EFA contents wasn't significantly difference in 4 periods(P>0.05).The breast and leg fatty acid contents was significantly difference in different chicken breeds(P<0.05). The monounsaturated fatty acids(MCFA) contents wasn't significantly difference in three chichen breeds(P>0.05),The difference of PUFA and EFA was significantly(P<0.05). The Wenchang chicken anfd Rugao chicken was lower than the local chicken breeds.The chicken fatty acids contents was different in different part of chicken. The breast SFA,PUFA and EFA contents was significantly higher than that of leg((P<0.05).The UFA and MUFA contents was significantly lower than that of leg(P<0.05). So the result showed that, The nutrition of breast and leg was different.The difference of male and female fatty acids content wasn't significantly(P>0.05).So the chang of fatty acid wasn't associated with sex.2.Determined the content of Intramuscular fa(tIMF)in breast and leg muscle in the three chicken groups.The IMF contents increased with age, The deposition rate of Anak chicken was faster than the local chicken breeds in breast,wasn't significantly difference in leg(P<0.05). But the deposition rate of leg was faster than breast.The IMF contents of female was a litter higher than that of male. But the difference wasn't significantly in Anak and Rugao chickens. (P>0.05).3.Inheritance analysis ofΔ6 fatty acid desaturate gene:there was some bp changed in the second intron and third extron,Detected three alleles,designated as A,B and C.The three alleles constructing the six genotypes,were distributed with Hard-Weinberg equilibrium among three chicken breeds .The genotypes,detected from 360 of total experimental population,influenced significantly polyunsayurated fatty acid and Essential fatty acid,The fatty traits of AB genotypes was higer than other genotypes.The results of this study suggest thatΔ6 fatty acid desaturate gene can be a linkage to it controll fatty acid trait and worthy of further studing as a molecular marker.4.Inheritance analysis ofΔ9 fatty acid desaturate gene:there was some bp changed in the second extron,Detected two alleles,designated as A and B.The two alleles constructing three genotypes,were distributed with Hard-Weinberg equilibrium among three chicken breeds .The genotypes,detected from 360 of total experimental population,influenced significantly polyunsayurated fatty acid and Essential fatty acid,The fatty traits of AB genotypes was higer than that of AA and BB.The results of this study suggest thatΔ9 fatty acid desaturate gene can be a linkage to controlled fatty acid trait and worthy of further studing as a molecular marker.
