PCR-RFLP and PCR-SSCP technologies were applied to detect the polymorphisms of FUT1 M307 gene, Mx1 gene, DQB gene, DQA gene, GH gene and ESR gene in Sutai pigs. The associations between these polymorphisms and growth, development and productive traits were analyzed using the SAS and SPSS general linear model(GLM). The purpose of this study was to facilitate the progress of the best-hybridized combination of specialized strain breeding by using marker assisted selection, and provide theoretic foundation for further cultivation on Sutai pigs. Results were obtained as follows:(1) The PCR products of the M307 of FUT1 gene were 161bp long, and three genotypes(AA, AG and GG)were shown digested by Hin6Ⅰ, the genotypic frequencies of FUT1 locus are 0.235(AA),0.609(AG)and 0.156(GG) respectively. it's allelic frequencies are 0.540(A)and 0.460(G)detected.This locus was not at Hardy-Weinberg equilibration(P<0.01)in Sutai pigs.(2) The list square means linear general model analysis results showed: There were significant association between genotypes and 35-day body weight, body length, body height, rear quarters width.The 35-day body weigh of individuals with AA genotype were significantly highe(rP<0.01) than those of individuals with GG, and AG genotype was significant highe(rP<0.05 )than that of GG genotype, and the values of other traits of individuals with genotype AA were significant higher than that with genotype AG and GG(P<0.05), but the backfat of AA genotype was lower than that of GG , and AG genotype.(3) Polymorphisms of Mx1 gene, DQB gene and DQA gene in Sutai pigs were analyzed with the method of PCR-RFLP and PCR-SSCP. The results showed: the gene loci of Mx1 and DQB-RsaⅠhad high polyomorphism, and the number of allele was three, and genotype was five. CC genotype was not found in Mx1 gene, and BC genotype was not found in DQB-RsaⅠgene, the allele A at Mx1 and DQB-RsaⅠgene loci occupied absolute predominance in terms of number. The gene loci of DQB- HaeⅢand DQA both had three genotypes, the allele D and C at DQB-HaeⅢand DQA occupied absolute predominance. We firstly detected a G/A mutation in the DQA exon 4 gene loci, and this mutation resulted in an amino and acid substitution, Ala→Thr.The genotype of DD was whidtype, and CC genotype was mutant. All the genes were at at Hardy-Weinberg equilibrationin Sutai pigs except DQB-RsaⅠg ene loci.(4) The relationship between the four gene locus and reproduction traits was analyzed.The results showed: According to Mx1, the growth performances (birth weight, 35 days body weight)of individuals with BC genotype of was a little higher than other genotypes, but there were no significant difference among them(P >0.05). As for the other three gene locus, the pigs with genotype AC, DE and DD were separately significantly higher than those with genotype AA, DD and CC (P<0.01).(5) The genotypes of Sutai pigs at GH locus were detected by PCR-RFLP. The PCR products were cut by ApaⅠ. The results showed: The frequency of the C allele was 0.821, apparently, the C allele is predominant. Meanwhile, the effects of different genotypes on some important growth traits involving the birth weight, 30 days body weight, 4-month body weight, 6-month body weight were analyzed. CC genotype is the highest expression of growth performance in different phases.(6) Polymorphisms of ESR gene in Sutai pigs were analyzed with the method of PCR-RFLPs. The relationship between ESR gene and reproduction traits was analyzed. The results showed that there were genotypes AA, AB and BB in Sutai pigs, the frequencies of the ESR genotypes were 0.089, 0.570, 0.341 respectively. The total reproduction traits of three PvuⅡPolymorphisms of ESR gene showed the tendency of AA>AB>BB. In first parity, the Number Born Alive(NBA)and Birth Litter Weight(BLW)had significant difference between AA and AB genotype as well as between AA and BB genotype(P<0.05).In later parities, the BB genotype were higher than AA and AB genotype(P<0.05)in Weaning Weight of Litte(rWWL)and significantly higher than AA and AB genotype(P<0.01)in Total Number Born(TNB).There were significant difference between AA and BB genotyp(eP<0.05)in Number Weane(dNW).(7) Polymorphisms of the candidate genes in group with FUT1 Resistance gene and Sensitive gene were analyzed.The results showed: The polymorphism was almost the same as undivided group.(8)The distribution difference of the candidate genes in group with FUT1 Resistance gene and Sensitive gene was analyzed, the genotypic frequencies of the good genotypes of ESR gene and GH gene in group with FUT1 Resistance gene were a little higher than the groups'with Sensitive gene.(9) Used the relative materials and experimental method, introduced and made an analysis on the progress of breeding Sutai pigs with high power of produce, high disease tolerance and efficiency.
