Polymorphism Of The FUT1 And Mx1 Gene And Its Relationship With Production Performance In Sutai Pigs

The polymorphism ofα1-fucosytransferase(FUT1) and Mx1 gene in 15 pig breeds included Duroc, Yorkshire, Pietrain, Landrace, Wuzhishan pig, Meishan pig, Fengjing pig, Erhualian pig, Sutai pig, Lingao pig, Leping spotted pig, Wanan spotted pig, Tibetan pig, Rongchang pig and Asian wild boar were investigated by PCR-RFLP in this study, then the alleles and genotype frequencies of the two genes were determined. At the same time, two unrooted consensus trees were constructed using Neighbor-joining based on Nei's genetic distance, and the relationship between the two genes and the production performance in Sutai pigs were analyzed. The results showed:(1) The PCR products of the M307 of FUT1 gene were 161bp long, and three genotypes(AA, AG and GG) were shown digested by Hin6Ⅰ; The PCR products of the M857 of FUT1 gene were 174bp long, and only two genotypes (AG and GG) were shown digested by AciⅠ. The result of sequence which concluded two locus showed that a single neucleotide changeed from G to A (G/A mutation) in M307 and T to C in M857 of FUT1 gene.(2) The AA genotype of M307 of FUT1 gene was only detected in Duroc and Pietrain, however, the AA genotype was not detected in Yorkshire, Landrace, all Chinese native pig breeds, Asian wild boar and Sutai pigs. The Asian wild boar and most of Chinese native pig breeds showed GG genotype; In M857 locus, all the Chinese native pig breeds showed only genotype of GG except Lingao pig breeds.(3) The PCR products of the Mx1 gene were 290bp long, and six genotypes (AA, BB, CC, AB, AC and BC) were shown digested by Hin6Ⅰ;The sequences showed the same as Genbank in AA genotype, mutations of G/T, A/G, A/G and G/A in locus of 2065bp, 2098bp, 2105bp and 2124bp in BB genotype, 11bp deletion and mutation of A/G in locus of 2098bp in CC genotype.(4) Only two alleles of A and C were shown in four kinds of foreign pig breeds, and only two alleles of A and B were shown in all Chinese native pig breeds and Asian wild boar. All six genotypes were shown in Sutai pig.(5) According to the gene frequency of FUT1 and Mx1 gene in different pig breeds, based on Nei's genetic distance, two unrooted consensus trees were constructed using Neighbor-joining method. The results showed that, all native pig breeds were near with Asian wild boar, and all the foreign pig breeds were near with Sutai pig.(6) The result of the relationship between different genotypes of FUT1 gene and reproductive performances in Sutai pigs showed that the productive performances of AG genotype were higher than the GG genotype. The weaning number of AG genotype were significantly higher than the genotype of GG in the first parity(P<0.05), and the total number born of AG genotype were significantly higher than the genotype of GG in the late parity(P<0.05).(7) The result of the relationship between different genotypes of Mx1 gene and reproductive performances in Sutai pigs showed high difference between different genotypes in the late parity. The BC genotype were significantly higher than the genotype of AA, AB, AC and CC in the total number born, weigth at birth, weaning number and weight at ablactating; and also significantly higher than the genotype of AA, AC and CC in the number born alive.
