Elaeagnus angustifolia L.a genus of Elaeagnaceae,in which the condensed tannin were its main bioactive components.Objective to investigate an effective method of production of condensed tannin via Elaeagnus angustifolia,this paper takes a research on temporal and spatial variation of condensed tannin content and effect of exogenous induction on condensed tannin accumulation in Eaeagnus angustifolia seedling;the regulation of condensed tannin biosyntheses in callus culture of Eaeagnus angustifolia had also been studied,the result as follows:1,Dynamic variation of condensed tannin contents in Elaeagnus angustifolia seedling There was significant difference in condensed tannin content among the Elaeagnus angustifolia leaf in the different month.Condensed tannin content reached two peaks at July and October during one growing season,27.28 and 34.36 mg·g-1DW respectively.The order of condensed tannin content in the different part of Elaeagnus angustifolia was:bark>root>leaf>one-year-old branch.Humus soil was suitable for plant growth and condensed tannin accumulation;fertilization was unfavourable to condensed tannin accumulation and promoting plant growth greatly,the corresponding was adequate nitrogen application increased both plant growth and condensed tannin content of Elaeagnus angustifolia leaf.2,Effect of exogenous induction on condensed tannin accumulation in the leaf of Eaeagnus angustifolia seedling Salicylic acid and fatty acid could promote the condensed tannin accumulation in Eaeagnus angustifolia seedling.The condensed tannin content among the Elaeagnus angustifolia leaf and one-year-old branch reached maximum(50.87 and 20.01 mgg-1Dw) in the third day after the spaying with 400 mgL-1 SA solutions,which were 2.43 and 1.53 times respectively than those of control;The condensed tannin content in the Elaeagnus angustifolia bark reached maximum (50.87 and 20.01 mgg-1DW) in the second day after the spaying with 250 mgL-1 fatty acid solutions,which was 1.77 times than that of control.3,The establishment of regeneration systemThe callus was induced on the MS culture medium attaching 2,4-D 1.00 mgL-1 + BA 0.50 mgL-1 by Elaeagnus angustifolia young leaf as explants,the frequency of callus induction rate can be as high as 98.00%;The MS + BA 1.00 mgL-1 was the best medium for differentiation of adventitious shoots in callus culture;The buds developed roots on MS medium with 1.00 mgL-1 IBA and formed plantlets,92.00%of which survived when transplanted to soil. 4,Techniques to improving condensed tannin in callus.The best callus growth and the highest condensed tannin production was obtained under the culture on MS basal medium containing 1.00 mgL-1 + BA 0.50 mgL-1.The optimum time of subculture was 20 d,the content of condensed tannin in callus was 19.46 mg·g-1DW. Differentiation of callus cultures did not favour the accumulation of condensed tannin,UV-B radiation and tryptone had remarkable promoting effects on the accumulation of condensed tannin in the callus of Eaeagnus angustifolia.The content of condensed tannin in callus was up to 56.25 mg·g-1DW,which 2.89 times higher than than that of control. |
