| The dry seeds of Indigofera pseudotinctoria Mats were irradiated by 60-Cobalt gamma rays with the dosages of 150Gy,200Gy,250Gy,300Gy,350Gy,400Gy,450G,respectively.The radiosensitive and major biological effects at M1 generation were investigated.350 Gy was chosen to irradiate the dry seeds for breeding purpose, several kinds of leaf color mutants were isolated.The major characteristics, multiplication and cultivation techniques of elite colorful mutants of Indigofera pseudotinctoria Mats were studied,the strategies for the extension and application of colorful Indigofera pseudotinctoria Mats were discussed.Main results are as follows:1.Biological effects at M1 generation:Simulative effects were observed in the lower doses,significant inhibition in the higher doses,and close relationships of doses, germination rate,survival rate and seedling height.350 Gy were considered to the optimal doses based on survival rate and seedling height.Leaf chemic such as white-strip and partly yellow-edge leaves were observed in at M1 generation,and shrunk leaves,late delays of seed maturing and low seed rate were observed in the higher doses,however normal seed rates were observed in the doses of 150~250 Gy.2.Leaf color mutation:Ten kinds of stable and obvious leaf color mutations,i.e. deep yellow,yellow,light yellow,yellow-green,yellow-white,white leaf edge,white tiger,light purple were observed at M2 generation.The frequency of yellow type was about 0.5×10-4,and white type was about 0.1×10-4,light purple type was very low in 1×10-6.Three types of mutations such as yellow,light purple and yellow-white were relatively in good biological yield.3.Characteristics of colorful Indigofera pseudotinctoria Mats.The whole growth duration of colorful Indigofera pseudotinctoria Mats was relatively prolonged,and the number of tiller,floret and seed grains were obviously decreased.However,improved hot and cold tolerances were found in yellow mutant.4.Propagation and cultivation techniques.The elite colorful mutants of Indigofera pseudotinctoria Mats could be propagated by seed production,the grafting of branches and small root.5.Extension and application.To be large-scale commercialization of colorful Indigofera pseudotinctoria Mats,it is important to collaborate with company,to strength the protection of intellectual properties(IP),and establish the standard and profession production base. |
