In this paper,some important effective factors on induction of multiple shoots via cotyledon nodes of soybean were studied and transgenic systems of soybean via Agrobacterium-mediated transformation with high transformation rate were optimized.Reliable regeneration soybean cultivars susceptible to SMV were selected as receptors.Transgenic system including of seed disinfection,shoot induction medium,infection bacteria liquid and root induction were optimized,the results as follows:the best sterilization method was ethanol with mercuric chloride firstly, then sterile water washing;Effect of MSB media added extrally 2mg/L 6-BA+0.2mg/L IBA on multiple shoots was best;Activated bacteria with YEB resuspension could get best inoculation;Later selection was identified better than another.MSB of half sucrose and 2mg/L IBA+0.2mg/L 6-BA were best media for root induction.In this paper,pac1 gene was firstly transferd into soybean cultivars susceptible to SMV by Agrobacterium-mediated.8 transgenic plants were obtained with PCR positive.Southern Blot analysis showed that pacl gene had been integrated into genome of soybean.
