This study were to research the polymorphism of Ghrelin gene in Gaoyou ducks,Lianchen ducks,Putian black ducks,Jinding ducks,Shaoxing ducks,Youxian Partridge ducks, Beijing ducks,Jianchang ducks ,and associations between Ghrelin polymorphic loci and Production performance in gaoyou duck.The results were as follows:1 Three pairs of primers GE1,GE2 and GE3 from several primers designed were polymorphous based on the sequence of ducks'Ghrelin gene(GenBank No. EF613552).Three genotypes were respectively detected by polyacrylamide gel electrophoresis in PCR products of the three primers.2 The results indicated that there are a 9bp deletions(TC)at the site of 167 bp.Single nucleotide loci mutations were T→C at 431bp site for GE2. Single nucleotide loci mutations were A→G at 907bp site for GE3.3 The three SNP loci in Ghrelin gene of Gaoyou duck were in Hardy-Weinberg Equilibrium.4 The analysis results show:in intron 1 loci, Beijing duck is an broiler-type breed ,the frequency of A allele in which was significantly higher than that in other breeds.In intron 2 loci,the frequency of C allele in Shaoxing duck was significantly higher than that in other breeds,and the distribution of genotypes had significant difference from that in Beijing duck (P<0.05) ,had highly significant difference from that in the other breeds(P<0.01).In exon3 loci, the frequency of F allele in Liancheng duck was significantly higher than that in other breeds ,and the distribution of genotypes had significant difference from that in Putian black ducks (P<0.05),and had highly significant difference from that in the other breeds(P<0.01).5 The mutation site in intron 1 in which the 9bp deletions were significantly associated with several traits,so the mutation site had significant effect on the selection of egg producing in Gaoyou duck population(P<0.05).The mutation site in intron 2 in which T changed to C had significant effect on the selection of body weight at first laying in Gaoyou duck population(P<0.05).The mutation site in exon 3 in which A changed to G made Ala→Thr at 53 amino acids,and had significant effect on the selection of highest clutch days in Gaoyou duck population(P<0.05).
