Danofloxacin is a kind of fluoroquinolones exclusive which could be administered by many pathway used in veterinary medicine,and has better water-solubility.It also has wide antibacterial spectrum and its antimicrobial activity is powerful.Meanwhile its adverse effect is less,this drag has very good pharmacokinetics character and the cross tolerance with other antibacterials is less.So that has been used in veterinary carp to treat bacterial infection of some farming animals.With the wide development of intensive culture,the fish of Carp(Cyprinus carpio L.) has often been infected by bacteriosis which was a restrict factor for sustainable development.Finding some effective medicine will great benefit the cultivation of the fish.Danofloxacin has a better inhibitory action in vitro for Aeromonas hydrophila (Ah),that is a main pathogenic bacteria to causing bleed disease.However,the effects of danofloxacin on antioxidant function and immune function in Carp had not been studied by the numbers.To understand the reactiveness of potential toxic effect of danofloxacin on antioxidant and immune function,effects of danofloxacin on antioxidant and immune function in Carp was studied in this thesis.The result is:The acute toxicity test of danofloxacin in Carp was studied.The LD50(medial lethal dose) and 95%confidence of LD50 of danofloxacin in the carp was 1502.1040 mg·kg-1 and 1307.8863~1738.6013 mg·kg-1 by oral administration.Effects of danofloxacin on the toxicity of carp wre studied by oral administration for 20 days at dosage of 17 mg·kg-1,40 mg·kg-1and 80 mg·kg-1.The result showed that at 5th day,the SOD activity of 80 mg·kg-1 group increased significantly,other test groups hadn't made any obvious change,and at 10th and 15th day the activity in these three test groups all remained higher value than control group. The SOD activity of all test groups decreased at 20th day,but it was still higher than control group.The activity of SOD in plasma remained lower level by oral danofloxacin of dosage at 17 mg·kg-1,40 mg·kg-1 and 80 mg·kg-1 all the time.The SOD activity in erythrocyte was opposite to plasma.The activity of CAT in liver was induced earlier then inhibited.CAT activity in erythrocyte is higher than control group obviously all the time.The activity of Na+,K+-ATP in liver and erythrocyte hadn't made any chage(P>0.05).In gill,the activity of Na+,K+-ATP of all test groups was not significant compared with control group during the test,it was lower than control group in 80 mg·kg-1 at 20th day exceptly.The GSH-PX and GST activity in erythrocyte hadn't made any chage,the NO concentration in liver and plasma was same as GST activity in erythrocyte.The MDA concentration in liver and erythrocyte remained unchanged,but can raised up in plasma(P>0.05).Danofloxacin could decrease the content of lysozyme in liver and plasma,and 80 mg·kg-1 dosage was markly.The idellus liver and spleen was not changed in all test group compared with control group.The phagocytic of leucocyte and phenoloxidase activity in surme was inhibited by danofloxacin.The number of erythrocyte in test group all increased to control group,but the leukocyte number remained unchanged.
