Plant endophytic fungi has been in the plant microecological environment for a long term,establishing a stable and harmonious relation with the host,followed by abundant secondary metabolites. In recent years,studies have shown that secondary metabolites from the plant endophytic fungi generally have antibacterial activity,and many bioactive substances of that are new compound which have not been found before,which shows great economic significance for the development and utilization of drugs which has antibacterial activity.In this paper,the endophyte was separated from Dioscorea nipponica leaves of different region,and its fermentation were handled,initially selected endophytic fungi and the active site which have antibacterial activity.Finally,the N-butanol parts of their fermenters and medicinal Dioscorea nipponica were carried out with a comparative analysis of TLC and HPLC.In the experiments,Bacillus subtilis,E.coli,Staphylococcus aureus,Staphylococcus albus,Staphylococcus citreus,Micrococcus tetragenus,Salmonella typhi,Klebsielapneumoniae,proteus species,Pseudomonas aeruginosa,Shigella flexneri,Bacterium paratyphosum B for 12 kinds of pathogenic bacteria as antagonistic instructions,both fermentation broth and mycelium extracts from 52 Dioscorea nipponica endophytic fungi were screened for antibacterial activity.The results showed that mycelium extracts of three strains produced antibacterial activity.Hemolysis experiments of 52 endophytic fungi of Dioscorea nipponica fermentation broth and the N-butanol extract of mycelia indicated that the N-butanol extract of mycelia of 47 Endophytic fungi may contain the composition of hemolysis.The TLC analysis of the Fermenters of 52 endophytic fungi showed that the N-butanol extract of the mycelium of C37,C30,C50 and herbs had the same blue spots in the same location,which indicate that Strain C37,C30,C50 metabolism may have the same composition with the Indigenous medicine.After the sample solution of 52 endophytic fungi was in calibration by diosgenin standard for HPLC analysis,Integrating with HPLC and UV absorption spectrum,the result was that the extracts of C37,C30,C50 mycelium all had a peak in the vicinity of diosgenin,whose largest UV absorption were the same as diosgenin,so they may be similar to diosgenin composition,which can be identified by further isolation and analysis.
