Brassica campestris L. (2n=2X=AA=20) is one of the basic diploids of Brassica crops, and belongs to self-compability. Self-compatible and self-incompatible materials of Brassica campestris were used to prepare hybrid combinations. The corresponding groups of F2 and BC1F1 plants were gained. Seed-setting rate and index of the self-compatibility were calculated, interaction between pollen and stigma of self-compatible and self-incompatible materials were observed by fluorescence microscopy. The self-compatibility of Brassica campestris were observed or analysized by histology and embryology,genetics,SSR markers. The results showed as follows:1. Pollen and stigma interaction were espialled, the results suggested that self incompatible materials controlled self-pollination by occuring obvious callose reaction, little pollen which were bonded together on stigma, germinated pollen was little, and germination was late. Although no callose reaction were observed in self-compatible materials, at the same time, pollen rapidly germinated on stigma, pollen tubes quickly entered into stigma and stigma organization, the aboves performed unique characteristics of self-compatible embryology. The callose reaction in stigma papilla cells could be used as a quality mark for detecting the strength of self-incompatibility or self-affinity.2. Self-compatible datas of F2 groups were statistically analysed, all of F2 groups accorded with segregation ratio of 3:1. BC1F1 occurred traits separation from backcross of F1 and self-incompatible parent, the ratio of self-compatible and self-incompatible descendant was 1:1; BC1F1 partly presented as self-affinity from backcross of F1 and self-compatible parent, so the self-compatibility of Brassica campestris was controlled by a couple of dominant gene which managed simple traits genetics.3. Bulk Segregant Pmalysis(BSA) was used to screen SSR primers, 60 pairs of primers were gained, and 3 polymorphic SSR markers were achieved, they were BRMS-006,BRMS-005 and BRMS-51, which could amplify different bands from self-compatible and self-incompatible parents or pools. BRMS-006(170bp),BRMS-005(150bp) and BRMS-51(250bp) all linked with self-compatible genes of Brassica campestris according to F2 groups analysis, the genetic distance separately were: 5.92 cM,3.47 cM,3.35cM.
