The low-molecular-weight glutenin subunit (LMW-GS) gene diversity of 150 wild emmer wheat accessions from Israel was evaluated by a set of group-specific LMW-GS primers. The relationship between the gene diversity and ecogeographical background was analyzed. Based on the conserved sequences of the known LMW-GS gene, a primer set was constructed for the isolation of LMW-GS genes from 24 wild emmer wheat accessions. One hundred and thirteen novel LMW-GS genes were isolated. The single nucleotide polymorphisms (SNP) variations in LMW-GS gene sequences from wild emmer wheat were detected, and the evolutional fashions in each region of LMW-GS genes were also analyzed. The main results are described as following:1. Using seven specific primer sets, higher genetic variations of LMW-GS genes in 150 wild emmer wheat accessions were detected. LMW-GS gene based genetic distances and ecogeographical factors in wild emmer wheat were not significant associated.2. One hundred and thirteen LMW-GS gene sequences were isolated from 24 wild emmer wheat accessions. The identity of LMW-GS gene sequences were found to be affected by the types of the deduced N-terminal. The abundant SNP variations were found, and 1 or 2 SNPs per 10 nucleotides could be observed. Thirty six haplotypes were detected, among which 17 haplotypes contained only a single sequence and 26 haplotypes were found in a single population.3. The 5' flanking sequences, single peptide, N-terminal and C- terminal sequences of the 113 LMW-GS gene sequences have been divided into several clusters based on the alignment and phylogenetic analyses, which was in agreement with each other and the classification based on the sequences of coding regions.
