The colibacillosis in piglets which bring huge economic loss to stock farming was an acute and highly fatal infectious diseases caused by Escherichia coli,in which the yellow and white dysentery were the most harmful and had highest disease incidence in clinical. The traditional Chinese medicine had non-specific antibacterial activity,low residual toxicity,and to which animals ware not easy to have drug resistance.Furthermore,the treatment of yellow and white dysentery of piglets by traditional Chinese medicine had various ways,low cost,and high cure rate,so using traditional Chinese medicine to prevention and treat yellow and white scour in piglets has become a primary means in scientific research and clinical.The further study of trace elements in traditional Chinese medicine in recent years found that selenium had intimate contact with diarrhea.While the experiments of correlation among Pulsatilla Decoction which have been reported in the literature with its anti-bacterial,anti-inflammatory,anti-endotoxin and other aspects of the pharmacological effects,the selenium and free radicals in blood were lack of systematic reseach.Based on this,the experiment was comparatively studied on the inhibition of pathogenic E.coli in vitro of the Pulsatilla Decoction preparated in two different way,the content determination of selenium in the two decoction and each single drugs,the pharmacodynamics characteristic of the Pulsatilla Decoction and its influence on the SOD activity,MDA and selenium in blood in the pathological model of diarrhea in piglet caused by E.coli in vivo.The purpose of the study was to found the mechanism in modern pharmacology of Pulsatilla Decoction's prevention and treatment with the diarrhea in piglets caused by pathogenic E.coli.18 healthy 20-day-old piglets constructing the pathological model of diarrhea caused by E.coli firstly were randomly divided into six groups with each group triplicates,ⅠGroups:neomycin hydrochloride was fed 15mg/kg.b.w,Ⅱgroups:the premix of vitamin E and selenite was fed 0.3mg/kg.b.w,Ⅲgroups:hydrochloride neomycin 15mg/kg.b.w was fed in company with the premix of Vitamin E and Sodium selenite 0.3mg/kg.b.w,Ⅳgroups:Pulsatilla Decoction was fed 50mL/head;Ⅴgroups:non-delivery under pathological conditions;Ⅵgroups:healthy contrasting group. The SOD activity,MDA levels and selenium concentrations in blood at different times were determined by hydride generation-atomic fluorescence spectrometry and the kits of SOD and MDA,among which the lowest detection limit of selenium concentrations in blood was 0.5μg/L,and the scope of standard curve was 0.0~400.0μg/L.Then use the software of SPSS 13.0 to analyze the data,and the results showed that:(1) The diameter of inhibition zone of the Pulsatilla Decoction extracted together to Escherichia coli was 1.070cm,while the Pulsatilla Decoction extracted separately was 0.803cm.The MIC of the Pulsatilla Decoction extracted together to the Escherichia coli was 125mg/mL,while the Pulsatilla Decoction extracted separately was 250mg/mL.the results showed the Pulsatilla Decoction extracted together was better in inhibiting Escherichia coli.(2) The determination of selenium concentrations in different preparation of "Pulsatilla Decoction" and the decoction of each drugs showed:the decoction of cortex phellodendri>the decoction of Coptis chinensis>the decoction of cortex fraxini>the decoction of Chinese pulsatilla root;"Pulsatilla Decoction" extracted together>"Pulsatilla Decoction" extracted separately,which was coincide with the MIC and the diameter of inhibition to Escherichia coli.(3) The diarrhea piglets caused by Escherichia coli with the SOD activity significantly decreased and MDA levels increased,after treated,the SOD activity no significant changes and MDA level has been in a relatively high level in western medicine group(neomycin), The reason of which may be related to endotoxin-induced release in the course of antimicrobial treatment;while the SOD activity in the other treatment groups were went down firstly,then step up to normal.(4)The piglets were cured separately by Neomycin,Sodium Selenite+Vitamin E,Sodium Selenite+Vitamin E+Neomycin,Pulsatilla Decoction after infected with Escherichia coli,by contrasted with healthy and pathology groups.The results shows:The selenium concentration was obviously lower than the healthy group and every cure group in pathologic status.In four treatment prescription,the group of Sodium Selenite+Vitamin E +Neomycin was notable better than just Sodium Selenite or Neomycin with Vitamin E, while the group of Pulsatilla Decoction had no significant differences with it.The results above showed:(1)The incidence of diarrhea in piglets caused by Bacterium coli had correlation with the concentration of selenium in blood. (2)The adjustment of Selenium in blood had positive significance in the treatment of diarrhea in piglets caused by Escherichia coll.(3)The mechanism of the treatment of Pulsatillae Decoction to diarrhea in piglets caused by Escherichia coli.was not only the non-specific antibacterial activity reported by existing literature,but also the adjustment of Selenium in blood.
