An effective seedling technigue was developed by studying the effects of development degree and medium types of Phalaenopsis seed embryos obtained by artificial pollination,.After a series of researches on sterilization methods,medium types,hormone concentration and browning control with explants of flower stem,an effective method for rapid propagation was found by clustered shoots.With stem segments as explants,somatic embryos were induced successfully and develop to plantlets.The results were as follows:1.Aspetic culture of Phalaenopsis seeds embryosThe best harvesting time of seed pots was 110 to 120 days.The most suitable medium for seed germination was 1/2MS+AC1.0g·L^-1;The most suitable medium for seedling tooting was 1/2MS+AC1.0g·L^-1+Banana pulp60ml·L^-1.2.Rapid propagation of clustered shootsThe 2-3cm cutting segments of flower stalks with an axillary bud on each were soaked into 0.1%HgCl₂ for 15min after series of pretreatment.The aseptic plantlet rate could reach 90%.The most suitable medium for primary culture was MS or 1/2MS+6-BA3.0mg·L^-1;The most suitable medium for subculture was 1/2MS+6-BA7.5mg·L^-1+NAA1.0 mg·L^-1;The most suitable medium for rooting was 1/2MS+NAA1.0mg·L^-1+AC1.0g·L^-1+ Banana pulp 60ml·L^-1.The survival rate of transplant after exercising seedlings for 7 days was 90%.3.Study on somatic embryogenesisSomatic embryos or somatic embryogenic callus were maintained from flower stem segments by culturing on 1/2MS basal medium supplemented with TDZ or 6-BA and NAA;All of somatic embryos were single cell origin,which would gradually develop globular embryo,heart-shaped embryo,torpedo embryo and cotyledon embryo via somatic embryogenic callus.The whole development process was similar to zygotic embryo.The somatic embryos could develop to plantlets finally.
