"Health" is becoming the most popular concern. So there is far-reaching realistic significance and expansive foreground in developing functional products from botanical resources for human being's health through the studying on physiologically active ingredient in natural plants. Because of its safety, nontoxicity as well as nutritional and the pharmacological functions, anthocyanin is drawing more and more attention from food industry, medical industry and consumers, thus it has huge potential in application. In this case, the development and utilization of natura1 pigments, especially anthocyanin pigments from plants is becoming one of the research hotspot at present.Anthocyanin has been used in food industry, cosmetic industry and medical industry abroad. It also has capacious markets in our country. But there are some disadvantages in the production and use of it, such as simplex variety, high cost, and instability. So it is high time to empolder new resource which has business value and stability. Purple maize, because of its more content, less cost and stability, is becoming a good choice. The systemic study on purple maize pigment will be helpful not only to develop and utilize this resource more timely, reasonably, effectively and thoroughly, but also to enlarge the cultivation and improve the regional agriculture structure which results in the increase of farmers, income.Choosing a purple maize plant (Qi319/lx530) for raw material, we conducted a study to determine the best extraction condition, superficial purification and the physical and chemical properties, which provided a reference for the development in many ways of purple maize.Details about this study were as follows:1. The maximum absorption spectrum of the pigment was 519.9 nm at the range of visible-light, so it was a kind of anthocyanins.2. By means of L9(34) orthogonal test, we determined the orders: extractive proportion﹥extractive temperature﹥extractive time﹥extractive pH. The best condition of the extraction was as follows: proportion of plant and extractive solution was 1﹕40, extractive temperature was 40℃, extractive time was 1h, suitable pH was 4.0.3. The equation of the extractive series was: y=4.5526e-1.3556x, the extractive series: 2.4. The main physical and chemical property of the pigment was: The pigment solution was steady red in acidic medium; however, the color and theλmax of the pigment changed in alkaline medium. Fe3+ and Cu2+ let the red solution fade and the pigment showed high stability in glucose, sucrose and sunlight. It was sensitive to high temperature, Vitamin C, oxida (H2O2) and reductance (Na2SO3).5. The silica gel chromatography was used to purify the pigment. It was feasible to use the solution (adjusted to pH4 by formic acid) which was made up of alcohol and water whose volume ratio was 7﹕3 as the washe impregnant.6. It was possible to conjecture through UV analysis that the pigment of the purple maize was Cyanidin or Delphinidin, or the mixture of them. The basic structure of the pigment was:7. The average gain rate of the whole plant pigments was 10.2%. There will be an expansive foreground because of this high content.
