Wheat male sterile line with Ae.kotschyi cytoplasm keeps itself in the foreground compared with others, but the major disadvantage that prevents it from being widely used in the field seed production is that most restorer lines have a relatively lower and unstable restoring ability. The wheat variety Yumai 2, developed by our work team, was the steady restorer with the K-type cytoplasm by identifying for many years and had the character of high restoring ability. The further study showed that Yumai2 is not only carrying Rfv1 gene on 1BS, but also bringing a new restorer genes and linkage with SSR marker Xgwm526 of 2BL chromosome. Understanding the genetic mechanism of its fertility restoration is helpful to select elite restoring lines and hybrid wheat lines with high restoring ability. It can facilitate the selection of fine hybrid combination and promote the hybrid wheat widely used in field production.The experiment was carried using K-type cytoplasm male sterility(CMS)of Yumai3 that it does not induce haploid, maintenance line of Yumai3 and restorer line Yumai2 with carrying two pairs of restoring genes; The Near-isogenic lines(NILs)of fertility restorer genes were constructed. In order to improve to select breeding efficiency of the NILs, the genetic background of NILs was explored by agriculture character and microsatellite marker (SSR) methods. The aim is to obtain good genetic consistency NILs.The results were as following:1. We selected 66 pairs of SSR primers that have detailed information on the basis of former research to detect the genetic background of NILs with K-type Cytoplasm Male of restoring gene. The results suggested that SSR markers were the effective tools to evaluate the difference of genetic background in the NILs population. The polymorphisms mainly existed among the 17 NILs and were highly distributed between the 17 NILs and CMS 1ine. On the basis of similarity coefficient, the cluster analysis was carried using SPSS software. Six NILs were higher consistency with the male sterile line including N15, N17, N10, N7, N8, N16. 2. The agronomic traits and cluster analysis between NILs of restoring gene indicated that backcross generations and the recurrent parent showed a higher consistency in morphological traits using CMS line Yumai3 backcrossed continuously with NILs 4 generation. N1, N7, N14, N16, N15 had the little difference with the male sterile line Yumai3.3. Combination of molecular markers and agronomic traits demonstrated that NILs of N16, N7, N15 and male sterile line of Yumai3 had more consistency. These NILs laid a solid foundation for fine mapping of the restorer gene, functional analysis and gene cloning research.
