Wheat male sterile line with Ae.kotschyi cytoplasm keeps itself in the foreground compared with others,but the major disadvantage that prevents it from being widely used in field production is that many restorers have a relatively lower and unstable restoring ability.The genetic mechanism of its fertility restoration is helpful to select good restoring lines and hybrid wheat with high heterosis.A set of crosses between different sterile lines and restorer lines were made,and segregating populations were derived.The inheritance of fertility restorer genes was studied,and the restorer genes were mapped to chromosomes using molecular markers.The type of the restorer lines was detected using molecular markers also.The results were as follows:(1)Ten backcross populations were derived by using ten restorer lines and two sterile lines,K-type Yumai 3 and K-type Yujiao 1,to study the number of restorer genes.The results are indicated that one pair of major restorer gene was found in the restorer lines Yumai 49,Bainong 64,Yunong 97202,Zhengyou 6,Xinmai 9 and Yumai 66,two pairs in Yumai 2,L783 and Shanl60,and maybe three pairs in Zhengmai 9023.(2)The backcross populations K-type Yumai3//Yumai3/Yumai2 and K-type Yujiao 1//Yujiao 1/Yumai 2 were used to map the fertility restorer genes of Yumai 2. Xgwrn18,Xgwm374,BARC061,TAGLGAP and DuPw214 on chromosome 1BS were linked to one fertility restorer gene,and Xgwm526 and cfd267 on chromosome 2BL were linked to another fertility restorer gene.The distances between the fertility restorer genes and the markers were different in the two backcross populations,but their relative chromosomal location in the two populations was the same.(3)The SSR marker Xgwml 8 and rye genome DNA primer AF1/AF4 were used to detect 1BL/1RS translocation in 168 restorer lines.27 restorer lines carried 1BL/1RS translocation and the other 139 didn't.The detected results for 98.81 percent of restorer lines by the two markers were the same,and only a few lines were different. 属性不符...
