| In this study, we chosen Chinese parents of japonica rice as materials and used SSR markers to construct their DNA fingerprints. By comprehensive analyzing their DNA fingerprints, we selected SSR markers with good stability and high polymorphism to construct DNA fingerprints of japonica rice. We aimed to provide technical support to protection of new varieties of japonica rice and species identification of the authenticity and purity .Based on SSR fingerprint database that was successfully constructed; we systematically study genetic diversity and phylogenetic relationship of Chinese japonica rice. It will help us to fully understand and use genetic resources of japonica rice. And it will provide evidence for parent match and varieties improvement.The main topic of this thesis is as follows:1.A set of primer pairs was selected from ,which are possibly chosen National Institute of Standards, with considering the balanced distribution in chromosomes, stability, high PIC value, reasonable distribution. The primer pairs consist of 12 core primer pairs and 12 Expand primer pairs as a complement core primer. The primer pairs fit the construction of SSR fingerprint of japonica rice.2.A database was constucted that contain genotype data of 42 materials of japonica rice at the 24 primer loci. The genetic diversity analysis of japonica rice showed that these materials have low heterogeneity and genetic diversity.3.All materials were classified into 5 groups, at the level of genetic similarity coefficient 0.73, according to cluster analysis by an un-weighted pair-group average method with arithmetic mean.As expected,the result of cluster roughly coincided with the genetic relation based on pedigree analysis. |
